Objective To investigate the influence of kidney-type glutaminase(GLS1)on biological function and vasculogenic mimicry(VM)in hepatocellular carcinoma(HCC).Methods Cell proliferation assay,wound healing assay and invasion assay were used to investigate the influence of GLS1 in SMMC-7721 cell line on biological function of hepatocellular carcinoma.The three-dimensional cell culture technique was used to observe the structure changes of vasculogenic mimicry by using GLS1 inhibitor and western blot was used to detect the influence of GLS1 on the expression of vascular endothelial-cadherin(VE-cadherin)and vascular endothelial growth factor(VEGF)in VM-related protein of hepatocellular carcinoma.Results The cell proliferation assay showed that the inhibition of GLS1 activity can inhibit the proliferation of hepatocellular carcinoma cells in a time and concentration-dependent manner.The wound healing assay results showed the relative migration rate of experimental group was significantly lower than the control group after scratched 24h(P<0.05).Transwell invasion assay results showed that at 24 h after the cells seeded,the invasive cell numbers of experimental group was significantly lower than the control group(P<0.05).Three-dimensional cell culture results showed that in the control group,the SMMC-7721 cell line showed vasculogenic mimicry,but in the experimental group,the vasculogenic mimicry structure was destroyed.The control group vascular mimicry formed tubules and nodes more than the experimental group,the difference was statistically significant(P<0.05).Western blot results showed that the VE-cadherin and VEGF protein expression have no obvious change,we haven't found any significant difference of the VE-cadherin and VEGF protein expression(P>0.05).Conclusion GLS1 can promotes the proliferation,migration and invasion of hepatocellular carcinoma.Inhibition the activity of GLS1 can destroy the vasculogenic mimicry structure,suggested that GLS1 may be involved in the angiogenesis of hepatocellular carcinoma.However,further studies are needed to explored.
