Objective:Through comparing behavior change and bowel dysfunction at different time in PD rats,to explore relationship of bowel dysfunction and behavior change in Parkinson’s disease.By observing the changes of α-synuclein and CXCR4 expression in substantia nigra and colon of PD rats at different time points,and to explore the early diagnosis method of Parkinson’s disease.Methods: 64 healthy female SD rats were randomly divided into PD group and control group,respectively after 0,2 weeks,4 weeks,6 weeks collect the rats 2 hours total feces,calculate the proportion of water content in feces,at the same time through the autonomic activities experiment and suspension test to observe rats behavior change;By using immunohistochemistry,Western blot to test the exprssion change of alpha-synuclein and CXCR4 in the substantia nigra 、 colon at different time in PD rats.Results:2 hours total feces and water content: Compared with the control group,the total amount of feces and water content decreased(P <0.05)in the PD group within 2 hours.Behavior change test: Compared with the control group,there was no significant difference in the PD group at the 2nd week(P> 0.05),there was significant decrease in the 4th week(P <0.05).Immunohistochemistry: the PD group from 2 weeks,the average optical density of α-synuclein in substantia nigra and colon was significantly higher than that in control group(P <0.05).The average optical density of CXCR4 in substantia nigra was significantly higher than that in control group(P <0.05).The average optical density of CXCR4 in colon was significantly lower than that in control group(P <0.05).Western-blot test: the PD group from 2 weeks,the expression of α-synuclein in substantia nigra and colon were increased,showed a increasing trend;The expression of CXCR4 in substantia nigra were increased,showed a increasing trend;The expression of CXCR4 in colon were descended,showed a descending trend.Conclusion:1.The bowel dysfunction was earlier than behavior change in Parkinson’s disease.2.The CXCR4 expression are positively correlated with the alpha-synuclein expression in the substantia nigra,they are negatively correlated in the colon.
