Objective:To observe the SCORAD scores,safety indexes,recurrence rate,and adverse reactions before and after treatment in patients with chronic spleen deficiency and dryness due to atopic dermatitis,and to explore the clinical effect of spleen deficiency and blood stasis syndrome in the chronic phase of atopic dermatitis.After in vitro experiments,CCK8 method was used to detect the effect of different concentrations of Jianpi Yangxue Qufeng Fang on the proliferation or inhibition of dendritic cells,and q-PCR was used to detect the concentration of spleen and nourishing spleen and nourish the wind to the dendrites.The expression levels of TLR2,TLR4,My D88,and NF-k B in the squamous cell were studied to investigate its effect on the skin's immune barrier.Methods:1.Clinical research Through clinical randomized controlled trials,60 cases were included and randomly divided into treatment group(Jianpi Yangxue Qufeng Recipe group)and control group(loratadine group),30 cases in each group.The course of treatment was4 weeks and followed up for 2 months.Before and during treatment(week 2)and after treatment(week 4),SCORAD scores and efficacy,as well as safety,adverse reactions,and recurrence rates were observed.2.Experimental research1.1 Spleen nourishing and expelling wind recipe on dendritic cell growth.1.2 The Effect of Spleen-Nourishing,Blood-Activating and Qufeng Recipe on the Expression of TLR2,TLR4,My D88 and NF-k B in Dendritic Cells.Results:1.Clinical observation results1.1 The total effective rate of 4 weeks for treating spleen and nourishing spleen and promoting blood circulation was 81.48%.The total effective rate was 44.3 weeks afterloratadine treatment was 64.38%.There was a significant difference between the two groups(P<0.05).1.2 Spleen and blood nourishing prescriptions can reduce the SCORAD score of patients,which is statistically significant(P=0.00).1.3 Follow-up results: The recurrence rate of spleen-enriching,blood-breathing and anti-fatigue prescriptions was 27.78%,and the recurrence rate of loratadine group was45.45%,P<0.05,with statistical significance.1.4 Adverse events: There were no adverse events in both groups and the safety was good.2.Experimental results2.1 Jianpi Yangxue Qufeng Prescription concentration 0-50 ?g/m L has a proliferation-dependent effect on dendritic cells in a concentration-dependent manner and inhibits dendritic cells when administered at a concentration greater than 100?g/m L.The effect is therefore the greatest value added at 50 ?g/m L.2.2 The expression of TLR2,TLR4,My D88 and NF-k B protein in dendritic cells was significantly higher in the spleen-enriching and blood-chilling group than that in the control group(**P<0.01).The highest level of expression occurred at a dose of50?g/m L.Conclusion:1.Spleen nourishing and expelling wind can reduce the SCORAD score of patients,safe and effective.2.Jianpi Yangxue Qufeng Fang has a value-added effect on dendritic cells within a certain concentration range;3.Jianpi Yangxue Qufeng can promote the expression of TLR2,TLR4,My D88,NF-k B protein in dendritic cells;...
