Objectives: Investigate and analyse the natural variation of Forsythia suspense of different producing area,according to phenotypic variation will be divided into four kinds of type.The quality of different populations and the four kinds of fruit type of Forsythia suspensa were evaluated.The genetic diversity of different populations and the four kinds of fruit type were analysed by molecular marker technology in order to determin natural variation effect and select the dominant population and fruit type which has excellent quality and genetic stability.To lay the foundation for Forsythia suspensa new breed breeding and standardized planting of GAP.Methods:(1)Quality evaluation of Forsythia suspensa of different populations were carried out by alcohol soluble extractive method and high performance liquid chromatography according to <Pharmacopoeia of People's Republic of China >2015 edition,it showed that the environmental factors are the main factors leading to the variation of Forsythia suspense.(2)The 9 natural populations of Forsythia suspensa as materials,The fruit vertical diameter,transverse diameter,hundred kernel weight,drying rate and appearance description as the index to classify the phenotype of forsythia fruit.Investigate the four kinds of type of 26 populations of Forsythia suspensa to determine the distribution.(3)Different populations and different fruit type of Forsythia suspensa was evaluated by ethanol soluble extract and HPLC methods according to <Pharmacopoeia of People's Republic of China >2015 edition to explain the differences in quality between different fruit type of Forsythia suspense.(4)Genetic diversity analysis by ISSR and SSR molecular marker technology.In order to clarify the genetic differences between different groups and quality differences between different fruit type.Results:1.The quality evaluation of Forsythia suspense of different populationsThe ethanol soluble extract,forsythiaside A and phillyrin of Forsythia suspense in different populations were determined and found that there are some differences in different populations through the variance analysis.The content of phillyrin in Yangcheng County,Jincheng and Taiping town,Xixia were higher,respectively 1.13%,0.99%.The content of phillyrin in Nanzhao,Nanyang was lower,0.27%.In the analysis of forsythiaside A,we found that the content of forsythiaside A in Licheng County,Changzhi was higher,Luoyang Songxian relatively lower,6.44%.There was significant difference between the two and other areas.The content of ethanol soluble extract results showed that the Changzi County,Changzhi and Lushan County Pingdingshan were higher,respectively 50.82 and 49.94%,the content of ethanol soluble extractives in Luoning County,Luoyang is relatively lower,35.20%.The results showed that these areas were significantly different from other areas.The analysis method of grey pattern recognition showed that in a comprehensive analysis of 26 populations of Forsythia suspense,the comprehensive evaluation score by 0.2918~0.6590,the Forsythia suspense of Taiping Town,Xixia County,Nanyang with better quality,Luoyang Luoning comprehensive quality score was the lowest.2.The study on natural variation of Forsythia suspenseOn the basis of fruit vertical diameter,transverse diameter,hundred kernel weight,drying rate and appearance description will be divided into 4 kinds of fruit type of Forsythia suspense,were spindle type(FC),apex(JFC),elliptical spindle type(TY)and peach(LY).In 26 natural populations of Forsythia suspense,four kinds of type is widespread in all populations and is the main source of production of Forsythia suspense in all areas,the analysis showed that the four kinds of type accounted for 37.62%,28.71%,21.78% and 11.88%.3.The quality evaluation of different types of fruitsMeasured the content of ethanol soluble extract and phillyrin of Forsythia suspense of different populations.The analysis of variance showed that fruit types had no significant differences,there was significant difference between only LY fruit of forsythiaside A with FC and JFC,LY of forsythiaside A content is the highest,10.03%.The grey pattern recognition method was used for the comprehensive analysis of different fruit types.The order was LY > FC > TY > JFC.4.Genetic diversity analysis by ISSR of Forsythia suspenseThe Forsythia suspense were amplified by 13 ISSR primers screened,amplified 353 bands,the percentage of polymorphic species level was 100%.Nei'S gene diversity index h was 0.2523,Shannon diversity index I was 0.3940,the gene flow among populations was 1.0070,and the coefficient of genetic differentiation ratio Gst was0.3318.In the analysis of the different fruit,the percentage of polymorphic species level was 98.87%,Nei'S gene diversity index h is 0.2403,Shannon diversity index I was 0.3783,gene flow between populations was 18.3017,the coefficient of genetic differentiation ratio Gst was 0.0266.NTSYSpc software for cluster analysis of 25 populations,25 groups can be divided into 2 categories,7 groups of cluster analysis.The 195 copies of Forsythia samples were divided into 2 categories,5 groups among individuals.5.Genetic diversity analysis by SSR of Forsythia suspenseThe Forsythia suspense were amplified by 14 EST-SSR primers screened,370 clear bands were amplified.the percentage of polymorphic species level was 98.87%.Nei'S gene diversity index h was 0.2796,Shannon diversity index I was 0.4315,the gene flow among populations was 1.5765,and the coefficient of genetic differentiation ratio Gst was0.2408.In the analysis of the different fruit,the percentage of polymorphic species level was 98.11%,Nei'S gene diversity index h is 0.2147,Shannon diversity index I was 0.3469,gene flow between populations was 8.6375,the coefficient of genetic differentiation ratio Gst was 0.0547.NTSYSpc software for cluster analysis of 12 populations,12 groups can be divided into 2 categories,7 groups of cluster analysis.The 77 copies of Forsythia samples were divided into 2 categories,5 groups among individuals.In the analysis of principal coordinates,individuals can be divided into 8 groups.Populations have rich genetic diversity,there is no correlation between genetic distance and geographic distance between populations of Forsythia suspense,mainly affected by ecological factors and growth environment.It showed that in the natural growth state,Forsythia suspense affected by environmental factors and ecological factors effects,which lead to the differences in view of the diversity of table traits and quality.This experiment shows that the diversity of Forsythia suspense is mainly affected by the ecological factors and the morphological phenotype related to environmental factors.Integrating five parts of the experiment,LY fruit forsythia can be recommended for the dominant fruit type and Taihang and Fu niu two mountains are main producing areas for the dominant group.It has great significance for the standardized planting of several varieties of Forsythia suspense.
