Study On The Quality Evaluation Of Schisandrae Chinensis Fructus Based On Commercial Specification Grade And Effect Component Index

The quality of Traditional Chinese medicine(TCM)is closely related to the curative effect.However,the quality control and assessment methods were mainly based on separate the component and measure the content at present,try to control several chemical content simultaneously to control the quality of TCM.However,most of these different chemical components are not related to each other,and they are not closely related to their safety and effectiveness.There were difference contribution of the different components to the same effect,and the different efficacy,the effective active components may not consistent.So,only through determination of the content of one or several kinds of chemical components may not be accurate and comprehensive to evaluation the quality of TCM.To explore a correlation function of new quality appraise method and control the quality of TCM,make the study of TCM from the fragmentation evaluation control which was difficult to associate the effect to integration the control index which can associate the clinic efficacy.Based on the previous of study in our team,the Schisandrae Chinensis Fructus(SCF)was used as example in this study and try to build an integrated control method of quality for TCM:Effect Constituent Index(ECI)which can evaluate the quality of SCF comprehensive.The main contents of this study include:1.The Dlphi method was used through the expert,establish a screening model to eliminate the experts and samples which were not meet the requirements.Quantitative analysis of expert's experience,make the quality identification card,and the identification card was given to the new learners to verify the inheritance of experience by the result of accuracy.The results shows that the new learners can distinguish the different grade of SCF quickly and accurately after used the quality identification card,and means that the experiences of experts can be inherited quickly by quantified it.The chemical content such as total linnans,polysaccharides and polyphenol were analysed in different grade of SCF which were identified by experts.The result shows that to the different grade of SCF,the content of polysaccharides and polyphenol have a positive correlated with it quality grade but the total lignnas has a negative correlated.This experiment shows that the human experience assessment for TCM was scientific and inheritable.However,this kind of method was also boundedness to the different efficacy of TCM.2.To the different efficacy such as heptatoprotecctive and antineoplastic about the SCF,the effective active content were different.This study through the literature search about the main efficacy and chemical component at present and choose the heptatoprotecctive as evaluation index,the main chemical component include Schisandrol A(Sch A),Schisandrol B(Sch B),Schisantherin A(Schi A),Schisandrin A(Schisan A),Schisandrin B(Schisan B),Schisandrin C(Schisan C).The HPLC was used to measure the six kind of lignans in different batches of SCF and the result shows that the chemical were different during the different batches of SCF.The Sch B and Schisan C were higher in Lioaning province than others,the Schisan A and Schisan B were higher in Jining province but the Sch A and Schi A were higher in Heilongjiang province.The total of six lignans in the SCF were highest which were hosted in Jiling and the Liaoning were lower.3.The hepatoprotective activity of the six kinds of lignanas were measured and the results show that the activity of this six chemical component were different.The activity of Sch A was better and the Schisan A was lower.4.Based on the basis of our previous work and preliminary experimental results,Try to build the hepatoprotective effect constituent index of SCF,and calculate the effect constituent index about the hepatoprotective effects from different areas of SCF.The results shows that the ECI of SCF in Tonghua,Liaoning province was higher and the SCF in Fusong,Liaoning province was lower.The sample of SCF which the ECI highest and lowest were selected to be verified based on the experiment of vivo and vitro.The result shows that two batchs of SCF have a significant effects of hepatoprotective but there also have no significant between the two SCF.5.The six kinds of lignnas were used to make the network pharmacology analysis and the mouse serum were used to make the metabonomics analysis.In summary,no matter traditional sensory evaluation method or modern content determination method maybe can reflects the quality of TCM from a certain point of view,but also can't be fully associated the effect.As the center of system about the TCM quality evaluation,ECI as a new evaluation index which was build by the chemical constituents and bioactivities of comprehensive weighted index and it can solve the problem which the different evaluation index can get the different result about the quality of TCM.The ECI can relate the effect of TCM only through the determination of chemical component and it was not increase the difficult of operation.The ECI can make a reference about the study of integration about quality,efficiency and dosage of TCM.
