Based on the foundation of quality standards, extraction and purification process ofSemen Ziziphus spinosa and Fructus Schisandrae Chinensis medicinal herbs, the articlemainly focuses on Semen Ziziphus spinosa and Fructus Schisandrae Chinensis asobjects of study. Furthermore, it has studied on the Pharmacodynamics and Mechanismsof Sedative-hypnotic of Semen Ziziphus spinosa and Fructus Schisandrae Chinensiswith pharmacological experimental methods.Objective:①To observe the spontaneous motion and sleeping time of mice usingdifferent extract from Semen Ziziphus spinosa and Fructus Schisandrae Chinensis.②Toobserve the effects of extract from Semen Ziziphus spinosa and Fructus SchisandraeChinensis on the EEG and α wave in the insomnia rats caused byDL-P-Chlorophenylalanine(PCPA).③To observe the effects of extract from SemenZiziphus spinosa and Fructus Schisandrae Chinensis on the content of MonoamineNeurotransmitter,Amino acid Neurotransmitter and Oxidative stress factor in thehypothalamus of insomnia rats caused by DL-P-Chlorophenylalanine to investigatemechanism.Methods:①220mice were divided randomly into A group and B group, A group micewere injected with threshold dose of sodium pentobarbital, record the sleep time of mice.B group mice were placed in ZIL-2programmed spontaneous activity box, detection thespontaneous motion of mice.②The model of insomnia rats were established by PCPAintraperitoneal injection, after the modeling, all the therapeutic group were treated withcorresponding drug for one week, using Powerlab biological signal acquisition andprocessing system records rats EEG.③The model of insomnia rats were established byPCPA intraperitoneal injection, after the modeling, all the therapeutic group were treatedwith corresponding drug for one week. Observe the pathological changes of the ratshypothalamus, Spectrophotometric detect the content of NO, SOD and MDA in thehypothalamus, Elisa assay the content of GABA, Glu,5-HT, DA,NE and IL-1β in thehypothalamus, immunohistochemical detect GABA, Glu protein expression, RT-PCR detect GABAARα1and GABAARγ2expression.Results:①Compared with normal group, the mice spontaneous motion was obviouslyinhibited and sleeping time was obviously prolonged (P<0.05or P<0.01) in waterextract, alcohol extract and alcohol-water extract group, especially for thealcohol-water group.②Compared with model group, the frequency was obviouslyinhibited and the amplitude was elevated of α wave in the alcohol-water group(P<0.05or P<0.01).③Compared with model group, in the alcohol-water group the content of5-HT, IL-1β, GABA and SOD in the hypothalamus of rats increased obviously(P<0.05or P<0.01), while the content of DA, NE, NO and MDA decreased obviously (P<0.05or P<0.01).Conclusion: the extract from Semen Ziziphus spinosa and Fructus SchisandraeChinensis has obviously Sedative-hypnotic effect. Its mechanism is related to regulatethe content of Monoamine Neurotransmitter and Amino acid Neurotransmitter in thehypothalamus of rats and improve the body’s anti-lipid peroxidation.
