Objective:By comparing the effects of Xiaoqinglongtang and anti-OX40L mAb to both TNF-? inflammatory factor and OX40L costimulatory molecule,explore the effects of Xiaoqinglongtang to TSLP-OX40L-Th2/TNF-? signaling pathway.Then give further study to the Xiaoqinglongtang curative effect mechanism of cold asthma,to discuss whether its targets are finished by affecting OX40L.Prove the effectiveness of classical prescription by mordern biological technology,bring Xiaoqinglongtang's medical superiority into full play,promote intelligent use of medicine in clinical practise,and explore new ways for studies of asthma treatments.Materials and Methods:Fifty female BALB/c mices(SPF,6-8 weeks,weight 20-22g)were randomly divided into five groups:Xiaoqinglongtang group(group A,n= 10),anti-OX40L mAb group(group B,n=10),control mouse IgG group(group C,n= 10),asthmatic model group(group D,n=10),normal control group(group E,n= 10).After feeding all groups a week,except of group E,other mouses are sensitized by i.p.and H injection of OVA and Al(OH)3.Then mice of these four groups were made cold asthma model by challenging via the airway with aerosolized OVA,meanwhile,putting at a cold room,where there was an air-conditioning blowing cold wind.Every group was intervened by its own medicine at the designated time.Twelve hours after the final antigen challenge,mice were anesthetized and killed,and blood and lungs were collected.Then prepare serum,tissue homogenate and lung sections,and detect TNF-? in serum by enzyme-linked immunosorbent assay(ELASA),detect TSLP in lungs by Western blot,and evaluate the expression of OX40L in lungs by fluorescent immunohistology.Results:(1)The symptom and physical sign were more obvious in group C and D than that of group A and B,mice of group E had no obvious sympt-oms.(2)Lung sections stained by HE showed that the accumulation of infla-mmation cells and trachea stenosis were more obvious in group C and D than that of group A and B,the accumulation of inflammation cells and trachea stenosis in group E were not obvious.(3)Tatal number of either TNF-? or TSLP was improved in group D than that of group E.Tatal number of either TNF-? or TSLP was reduced in group A and B than that of group D.The above differences were both statistically significant(P<0.05).The difference between group A and B of total number of either TNF-? or TSLP had no statistical significance(P>0.05).(4)Luorescent immunohistology showed that the expression of OX40L in group A was " +?++",group B was"+",group C was "++?+++"group D was "++?+++" and group E was"-"Conclusions:Xiaoqinglongtang can lower the expression of OX40L in lungs of cold asthma mice to regulate TSLP-OX40L-Th2/TNF-? signaling pathway,then reduce the accumulation of inflammation cells in lungs.So Xiaoqinglongtang's targets to treat cold asthma may be realized by intervening the expression of OX40L.
