Experimental Research On The Effects Of Xiaoqinglongtang And Its Demolition Parts On ET And Eotaxin In Asthmatic Mice

Objects:To observe the effectiveness of Xiaoqinglongtang and its parts on he level of endothelin (ET) and eosinophil chemotactic factor (Eotaxin) in serum,and the pathological changes of lung tissue in asthma mice and its effects.From the level on endothelin (ET) and eosinophil chemotactic factor Eotaxin) in serum of mice in asthma attack,study of the decoction on these wo aspects can be proved clearly and thoroughly.By finding out the main )arts in changing the serum endothelin and eosinophil chemotactic mice of isthma attack,the rationality and scientific quality can be proved by the mechanism of asthmatic airway inflammation and high reaction of airway, and clarify the inside roles as the basis for the further clinical usage.Methods:60 healthy male KM mice(4-5weeks age,SPF level,20-25g weight) were randomly divided into six groups,10 for each group:normal group(GroupA),asthma group(GroupB),XQLT group(GroupC),WFHY group Group D),SLQY group(Group E),JB group(Group F).On the first.7th.14th lay of the experiment, mice from group B to group F had the abdominal cvity injections of egg albumin (OVA)/aluminum hydroxide gel suspension iquid 0.2 ml sensitization,and NS in group A.On the 15th day,all mice were ut in homemade plastic box to inhalation,2% egg albumin/NS 2 ml was ased in group B to group F to trigger asthma,with 2 ml NS in group A,(each group 30 min/d for 7d).Dosage:From the first day of induced asthma attacks, mice in C to F were given certain Chinese medicine granules by gavage half an hour before atomization (0.2 ml/10 g).After given medicine,the mice were weighted every 2 days and recorded the numbers for adjusting the dosage in time.Preserved specimens:In 12 hours after the atomization, speciments were made by eyeball-extracted and blood-drown.At the end of the experiment,by analysing the changes of ET and Eotaxin in serum, different influence of asthma attack mice in different groups were discussed. Meanwhile,the pathological tests of lung cells after HE staining were done n order to observe the situation of lung inflammatory cells infiltration and changes of the airway.Results:Compared with the blank control group,ET and Eotaxin increase in model group,WFHY group,QYLX group convergence was statistically significant,p< 0.05;Compared with model group,the whole party,WFHY group,JB group ET and Eotaxin decline was statistically significant,p< 0.05, QYLX group has no statistical significance,p> 0.05;Compared with the whole group,JB group,WFHY group was statistically significant,p< 0.05. That means in asthma,XQLT group,JB group,WFHY group can reduce ET and Eotaxin content in serum,SLQY group is not obvious;Compared with he XQLT group,JB group,WFHY group have difference in reducing the ET and Eotaxin content in serum.Lung tissue pathology biopsy showed:There is no inflammatory cells phenomenon in the lung tissue of blank control group, with normal structure,bronchial in smooth,complete and no epithelial exfoliated condition,cilia orderly,and no hyperemia.Model control group: here can be seen the infiltration in lung tissue and neutrophils and mononuclear lymphocyte mainly in submucous layer of bronchi,with a small amount of goblet cell hyperplasia,clear lung tissue hypermia,thicken rachea,hypermia in vessels,loss of bronchial cilia as adhesion,lodging and hecrosis.Compared with model group,it has an alleviation in Chinese medicine groups in the condition of pulmonary inflammatory infiltrates and hicken,bronchial wall.However,the most effective method is showed in the XQLT group,and the thick bronchial wall exists among the rest of the groups with no differentiation in lung pathology.Conclusion: ① The partly function of Xiaoqinglong tang in the treatm ent of asthma may be inhibiting ET level and Eotaxin activity in seru m and lung tissue of asthma mice.②In all intervention groups,JB grou p is most close to XQLT group, WFHY group also play an important role,③The effect drugs of SLQY group is not obvious, the treatment effect may be from other mechanism to asthma,but also is an integral part of the whole party.④Confirmed that Xiaoqinglongtang had the full and rigorous compatibility and drug combination is reasonable once again.
