Objective: Our previous studies have shown that miR-711 can inhibit gastric cancer cells occur EMT,but the specific mechanism is unknown,this topic on this basis,to explore the correlation between miR-711 and Sp1,further defined miR-711 regulation of gastric cancer cells mechanism of EMT.Methods: 1)Cultivation of gastric cancer cell line MGC-803 and SGC-7901 cells,using liposome lip2000 instantaneous turning technology,expression?inhibition of expression of miR-711,48 h after observation of transfection efficiency,Western Blot detection Sp1 protein expression.2)Construction of plasmid Sp1 interference,transfection gastric cancer cell line MGC-803 and SGC-7901,Western Blot detection Sp1 protein and EMT epithelial markers E-cadherin,mesenchymal markers vimentin protein expression quantity changes.Results:1)Transfection after 48 h,transfection efficiency of about 60%;Western Blot results,the expression of miR-711,reduce the amount of relative to express the Sp1 protein(p<0.05);Inhibition of expression miR-711,relative expression of Sp1 protein increased(p<0.05).2)After the plasmid transfection Sp1 interference,the gastric cancer cell line MGC-803 and SGC-7901 Sp1 protein expression of relative quantity are reduced,mesenchymal markers relative expressed vimentin were reduced,epithelial markers E-cadherin expression quantity are increasing(p<0.05).Conclusion:Mi R-711 can reduce the expression of Sp1 inhibiting gastric cancer cell lines of EMT.
