Objective:This study aims to analyze the allelic and haplotypic diversities of 15Autosomal and 10 Y-chromosomal short tandem repeat?Y-STR?loci in Chinese Southern Shaanxi Han population.The results of this study will be useful for enriching the genetic data of Chinese populations and will provide a reference for forensic cases.The genetic relationships between Southern Shaanxi Han and other groups were studied based on the genetic data in published researches.Method:214 healthy,unrelated individuals of the Southern Shaanxi Han population were investigated by fluorescence labeled Expressmarker 16+10Y?EX16+10Y?multiplex PCR system;the PCR amplification products were detected by PRISM ABI 3130 genetic analyzer.The genotyping data of 17other groups were downloaded from the database.The locus-by-locus Fst was calculated using the method of analysis of molecular variance by the software ARLEQUIN v3.1;the DA distances were calculated using the DISPAN program;the phylogenetic trees were obtained using the software MEGA v5 with the unweighted pair-group method with arithmetic means and the software PHYLIP v3.6 by the neighbor-joining method;the principal component analysis was conducted with the software MVSP.Results:The typing data of 15 Autosomal STR and 10 Y-STR for the Southern Shaanxi Han population were obtained.We observed a total of 150 alleles at 15 autosomal STR loci in the population with the values of allelic diversities ranging from 0.0023 to 0.5210,and the combined power of discrimination and exclusion for the 15 autosomal STR loci were0.999999999999999998869 and 0.999998491,respectively.For the 10 Y-chromosome STR loci,totally 100 different haplotypes were obtained,in which 94 were unique,the discriminatory capacity and haplotype diversity value of the 10 Y-STR loci were 0.9259and 0.998269,respectively.We analyzed linkage disequilibrium,neighbor-joining tree,principal component analysis and DA distances,which indicated that the studied Southern Shaanxi Han population had a close relationship with Eastern Han population.Moroccan,Croatian and Serbian were relatively far away from Southern Shaanxi Han.Conclusions:Through this study,the results demonstrated a high genetic diversity of the 25 STR loci in the population for both forensic applications and population genetic studies,and enriched the ethnical gene information resources for the Southern Shaanxi Han population.
