Objective: To detect the expression level and expression pattern of B7-H3 at different stages of gastric cancer,To determine the relationship between B7-H3 expression and clinicopathological features of gastric cancer;To detect the expression level of B7-H3 in peripheral blood and corresponding tumor tissues and further explore the expression of B7-H3 in gastric cancer and its potential clinical value.Methods: Gastroscopy and surgical resection specimens were collected,including 201 cases superficial gastritis,atrophic gastritis,low grade intraepithelial neoplasia,high-grade intraepithelial neoplasia,and gastric cancer.The expression levels of B7-H3 in various tissues were detected by immunohistochemistry staining and Real-Time PCR.The levels of soluble B7-H3 in plasma were detected by ELISA,while the expression levels of B7-H3 on CD14+ cells in the peripheral blood were detected by flow cytometry.The correlation between B7-H3 expression and clinicopathological features of gastric cancer were analyzed.Results: B7-H3 was almost not expressed in the superficial gastritis and atrophic gastritis.B7-H3 was expressed in the intraepithelial neoplasia,and the expression of B7-H3 increased with the grades of neoplasia.In gastric cancer,B7-H3 was widely expressed both in tumor tissue and interstitial tissue and its expression increased significantly.In gastric cancer,the expression of B7-H3 positively correlated with tumor size and TNM staging(p <0.05),but not correlated with gender,age,depth of invasion,lymph node metastasis,distant metastasis,and vaginal tumor thrombus(P> 0.05).The expression of B7-H3 mRNA in tumor tissue was significantly higher than that in normal tissues(P <0.05).The expression of CD14 + B7-H3 + was unchanged after surgery.The expression of plasma B7-H3 was associated with the expression of B7-H3 in tumor tissue.The expression of plasma B7-H3 was positively corrected with the number of Ki67 expressed cells in patients.Conclusion: The expression of costimulatory molecule B7-H3 in the evolution of gastric cancer suggested that B7-H3 plays an important role in the progression of gastric cancer.The expression level of plasma B7-H3 was positively correlated with cell proliferation.B7-H3,which was highly expressed in tumor tissue,can be secreted into serum.The plasma B7-H3 is expected to be a biological marker of gastric cancer and may have clinical applications.
