Objective:Study the effects of IncRNA MEG3 in prostate cancer cell lines.Quantitative RT-PCR assay to detect the expression of MEG3 in urine and to know its value for early diagnosis and prognostic in prostate cancer.Method:MEG3 was transfected into LNCaP and DU145 cell lines in vitro.Effects of MEG3 were evaluated by CCK-8 assay,colony formation assay.MEG3 and NC was transfected into PC3 cell line.Transwell chamber assay was performed to observe on its role of migration and invasion,respectively.Otherwise,a total of 115 patients scheduled for biopsy was registered in this study,including 53 positive patients and 62 negative patients.Post DRE urine sample were collected and treated immediately.Total RNA was extracted using Trizol reagent,reverse transcript into cDNA.MEG3 was determined by quantitative RT-PCR.The expression of MEG3 was compared with Gleason score and tPSA level.And we evaluate its clinical value for early diagnosis.Results:CCK-8 assay showed that upregulated of MEG3 can inhibit the proliferation of prostate cancer cells LNCaP and DU 145.Colony formation in vitro confirmed that MEG3 could inhibit the number and size of the clone of both prostate cancer cells LNCaP and DU145.Transwell chamber assay demonstrated MEG3 decreased the ability of cellular migration and invasion.The expression of MEG3 was lower in prostate cancer group than in hyperplasia group.But there is no correlation between Gleason score and tPSA.Conclusion:MEG3 plays as a tumor suppressor gene,and it can inhibit the proliferation,migration and invasion of prostate cancer cells.There is certain significance during MEG3 test in urine for early diagnosis of prostate cancer.
