ObjectiveTo study the effect of mi R-7-5p in energy metabolism pathway and biological function in prostate cancer cell lines,and further explore the mechanism of mi R-7-5p in prostate cancer cellsMethods:LNCaP?DU145?PC3 cell line were transfected with mi R-7-5p lentivirus and mimics.CCK-8 experiment and clone formation test cell proliferation ability.In addition testing the glucose and lactate levels,ELISA kit were used to exam G6 PD NADPH ?CITRATE enzymes levels and it was found by database that mir-7 and MEG3 binding each other and the level of mir-7-5p were correlated with MEG3 expression,transfecting mir-7-5p inhibitor into LNCa P DU145 PC3 cell line,we got different result in CCK-8 experiment and clone assay The downstream target genes chosen by RNA-sequence and further clarified the mechanism and signal pathway of glucose metabolism in prostate cancer.ResultsThe CCK-8 experiment showed that the proliferation ability of LNCa P,DU145 and PC3 cells was inhibited after mir-7-5p lentivirus and mimics transfected.In vitro cloning experiments showed that the clone formation ability of LNCa P,DU145 and PC3 of prostate cancer cells was also inhibited.And it also inhibited the energy metabolism in prostate cancer cells,the consumption of glucose decreased obviously,and compared to the control group,the lactic acid production was significantly reduced,and the content of enzymes like G6 PD,NADPH,CITRATE were significantly lower than the control group,by RNA pull down and RNA-seq PIK3CD?IL6R downstream target were identified Further explore the signaling pathway related to glucose metabolism and understand the mechanism of mir-7-5p in prostate cancer.Conclusionmi R-7-5p play as a tumor suppressor gene in prostate cancer,mi R-7-5p can significantly inhibit prostate cancer cells glucose uptake,mir-7-5p can combined with MEG3.inhibit the proliferation and cloning in prostate cancer cells the anticancer effect is clear..
