Study On The Isolation,Purification And The Anti-myocardial Ischemia Reperfusion Injury Activities Of The Polypeptides From Velvet Antler

Velvet antler,which is collected from the deer horn of the male sika deer(Cervus nippon Temminck var.mantchuricus Swinhoe)or red deer(Cervus elaphus L.),is one of the famous and valuable traditional Chinese medicine.It's been reported to have effects on improving immune system and cardio vascular systems,providing vitality and anti-aging properties,and enhancing sexual function.It's also been introduced to clinical neurosis,anemia,impotence,seminal emission,premature ejaculation,and cardiac symptoms treatments.Velvet antler has numerous bioactive substances,including protein,polypeptide,polysaccharide and polyamines.The crude polypeptides from velvet antler have been demonstrated to exert protective effects against acute ischemic myocardial injury(AIMI)in rats.In the present study,we isolated and purified a single polypeptide from the above mentioned crude polypeptides of velvet antler.The effect and underlying mechanism of this single polypeptide on myocardial ischemia reperfusion injury(MI/RI)in rats were investigated.Myocardial ischemia reperfusion injury occured after a critical period of coronary occlusion,and is a clinical damage to the myocardic tissue.Although the restoration of blood flow to the jeopardized myocardial area is a prerequisite for myocardium salvage,reperfusion itself may lead to the accelerated processes of ischemia and additional injury.The phenomenon is termed “Ischemia/Reperfusion injury(I/R)”.Since the reperfusion injury occurs during the treatment of myocardial infarction,attenuation of the injury extent while blood flow is being restored becomes a very important subject.In the past decades,more and more complementary treatments like folk remedies were used to decrease the injury.A lot of traditional Chinese medicines have been reported to have positive effects on the control of reperfusion injury.There are many chemiacal substances in velvet antler,We isolated and purified the polypeptides from velvet antler,and then test the biological activity.,we used themethods of extration such as centrafugation,dialysis and so on.When we purified them,we adopted modern biological isolation technique of gel filtration chromatograph,ion-exchange chromatograph,reverse C18 chromatograph ect.We tested the physical and chemical properties through the techniques of folin-phenol,HPLC and SDS-PAGE.We obtained the polypeptides from velvet antler by modern biological isolation technique.The molecular weight is estimated about 6500 Da through SDS-PAGE.The polypeptide VAP exerts significant cardioprotective effects against acute ischemic myocardial Ischemia/Reperfusion injury in rats,VAP also significantly decreased levels of CK,LDH,AST,increase SOD,CAT,GSH-Px activity in myocardial tissue in MI rats.
