Velvet antler is a kind of rarely tradional medicine,it has extensive effect on curing disease.Velvet kidney soup is the use of velvet and other Chinese herbal medicine brewed from the preparation,with strong bones and other effects.Velvet antler 32 polypeptide of sika deer is a kind of peptide extracted from fresh antler,which has obvious actives on promoting proliferation and inducing differentiation in MSCs.Both of ADSCs and MSCs come from mesoderm,which are high-profile in the research of bone injury repair and anti-tumor.Therefore,this experiment uses ADSCs as experimental cells.The aim of this research was to investigate the effects of antler powder and Velvet antler 32 polypeptide on the proliferation,migration and adhesion of ADSCs in mice and 293 T,and to further explore its molecular mechanism.In this research,we observed the effects of antler powder and velvet antler 32 polypeptide for proliferation,migration and adhesive force of ADSCs and 293 T.The effects of Velvet antler 32 polypeptide on the transformation of mouse ADSCs into osteoblasts and the changes of TGF-β1,Smad1,Smad2,Smad3 and ALP expression were also observed during the transformation.MTT shows that antler powder and velvet antler polypeptide has obvious effect to promote ADSCs proliferation in a dose and time dependent manner to some extent.Cell wound healing assay shows the width of ADSCs and 293 T in the antler powde was larger than that of the control group,concentration of velvet antler polypeptide with 1 μg/ml,the scratch width is narrower than control group,and with the concentration of 50μg/ml or 5μg/ml,the scratch width is bigger than control group.Trypsinization assay shows that as the increase of concentration with antler powder and velvet antler polypeptide,the cell digestion time shorten significantly and the cell adhesive force reduce gradually.Mineralization experiments showed that the number of calcium nodules in the experimental group was higher than that in the control group,and the expression of TGF-β1 was increased and the expression of ALP was decreased,Smad1 protein expression decreased,while Smad2 and Smad3 protein expression increased during the transformation.This study reveals that antler powder can promote the proliferation of ADSCs,inhibit the migration of cells and decrease the cell adhesion,and it is not significant to 293 T of proliferation,and can inhibit cell migration.Velvet antler polypeptide can promote proliferation of ADSCs and reduce the cell adhesive force,what’s more,a low concentration of velvet antler polypeptide can promote cell migration,higher concentration can inhibit cell migration.Velvet antler polypeptides can promote the transformation of ADSCs into osteoblasts,and promote the expression of TGF-β1,Smad2 and Smad3,inhibit the expression of Smad1 and ALP.It is suggested that the antler peptide can be used to accelerate cell proliferation and reduce its adhesion in specific areas,and to promote its transfer to damaged parts,and to explore its mechanism for the treatment of bone tumor and injury.The results can provide a theoretical basis for the further clinical application of velvet antler polypeptide extracts.
