Died of stomach cancer is the world's second-largest malignant tumor and gastric adenocarcinoma and accounted for more than 95%.In China,the gastric cancer with high morbidity and mortality and very low five-year survival rate for patients,gastric adenocarcinoma is a serious public health problem in China,there is an urgent need to address the issue.This study explores miR-199b-5p relationship between stem cell markers Nanog and the gastric cancer stem cell marker CD44,and sequencing of gastric adenocarcinoma cancer stem cell and cancer cell screening differential expression of miRNA,aims to provide an in-depth understanding of role of miRNA on cancer,for the treatment of gastric cancer with new research ideas and molecular targets.Our sequencing and bioinformatics by tumor cells,miR-199b-5p target sequences are predicted at Nanog and CD44,3'UTR region,may inhibit the expression of Nanog and CD44.Experimental part mainly through expression in gastric adenocarcinoma cell line MGC803 miR-199b-5p to observe the Nanog and CD44 gene expression and effect of protein translation.Results show that the miR-199b-5p has no effect on Nanog,expression of CD44 expression in MGC803 cells in the miR-199b-5p decreased,these results suggest miR-199b-5p inhibits the expression of CD44 gene.By overexpression of miR-199b-5p in the MGC803 cells,and observe the effect of miR-199b-5p on gastric adenocarcinoma cell biological function.Studies have found that miR-199b-5p increases significantly inhibit gastric adenocarcinoma cells MGC803 proliferation,migration,invasion and resistance level.These experiments show that miR-199b-5p acts as a tumor suppressor gene,inhibiting this process,migration,invasion and resistance.miR-199b-5p cell biology may be associated with the Suppression of CD44.Our conclusion is that miR-199b-5p is a potential cancer stem cell marker CD44 have regulation of miRNA,by inhibiting the activities of CD44 to suppress a number of tumor cell biology.For gastric adenocarcinoma cancer stem cell marker CD44 research broadens the base of the signal path,also for tumor diagnosis and treatment provided to a new molecular target.However,it is worth noting is,miR-199b-5p in our experimental results show that can inhibit the occurrence of cancer and development,but we see results by sequencing,expression in cancer stem cells is much higher than the tumor cells.We can only guess at the self-help or ceRNA with cell against tumor control.Further studies need to be confirmed.
