Objective This article is aimed to explore the relationship with salicylic acid and metastasis of gastric cancer and its possible mechanisms.Methods the rank concentrations of salicylic acid were used for dealing the gastric cancer cell MGC-803 for selecting the suitable concentration.The biological characteristics of cell growth,adhesion,migration and invasion of gastric cancer cells were investigated with MTS assay,cell adhesion assay,Transwell invasion assay under the 0.5mmol/L and 1mmol/L of salicylic acid.RT-q PCR is performed to test the level of m RNA in cell adhesion molecule CD44,CDH1,CTNNA1,FN,ITGA9,Lamin,Lamin B1 and Lamin C1.Cell adhesion molecule CD44,CTNNA1,FN1 and Lamin B1 protein expression is used to test by Western-blot.Results our results demonstrate that 0.5mmol/L and 1mmol/L of salicylic acid dealing with 24 hours are suitable for our studying based on the rank concentration of salicylic acid.We show the ratio of cell growth inhibition of 0.5mmol/L and 1mmol/L salicylic acid was(32±4.5)% and(38±4.1)%.Compared with the control(121.00±2.65),0.5mmol/L(88.00± 1.53)and 1mmol/L(44.00±2.00)salicylic acid decrease the number of cell adhesion.The migration assay suggests the ration of cell migration of 0.5mmol/L and 1mmol/L salicylic acid was(10.00±3.85%)and(12.00±3.61)%.We find 0.5mmol/L and 1mmol/L salicylic acid has the lower OD(0.18±0.00,0.15±0.00)of invasion than the control(0.22±0.01)in gastric cancer cell.Besides salicylic acid downgrades the expression of CD44,CTNNA1,FN1,ITGA9,Lamin,Lamin B1 and Lamin C1,however,upgrading the expression of CDH1.Last,the data displays that the level of protein expression of CD44,CTNNA1,FN1 and Lamin B1 were decreased after the salicylic acid treatment.(P<0.05)Conclusion 1.Salicylic acid inhibits the cell proliferation,adhesion,migration and invasion in gastric cancer cell.2.Salicylic acid downgrades the expression of CD44,CTNNA1,FN1,ITGA9,Lamin,Lamin B1 and Lamin C1,however,upgrading the expression of CDH1.Cell adhesion molecular is related with the metastasis of tumor,meanwhile the epithelial-mesenchymal transition possible is another mechanism contributing to the transfer of gastric cancer cell.
