Objective: To establish the oxaliplatin resistant human SGC-7901 gastric cancer cell and Study on its molecular biological characteristics and relationship between epithelial mesenchymal transition(EMT)Methods:(1)Induced human SGC-7901 gastric cancer cells to become resistant cells with the increasing usage of low concentration of oxaliplatin;(2)Compared the morphological difference between oxaliplatin resistant human SGC-7901 gastric cancer cell and parental cells with inverted phase contrast microscope;(3)Compared the Proliferative capacity at the same time and drug sensitivity of the two groups of cells with MTT;(4)Compared the difference of metastasis and invasion between the two groups of cells with Transwell experiment;(5)Compared differential expression of vimentin and epithelial cell adhesion factor(E-cadherin)between the two groups of cells with Immunofluorescence assay and Western blot.Results:(1)Compared with the parental human gastric cancer cells SGC-7901,the phenotype of oxaliplatin resistant human SGC-7901 gastric cancer cell Changed to mesenchymal cell phenotype;(2)Compared with the parental human gastric cancer cells SGC-7901,the proliferation in the same time of oxaliplatin resistant human SGC-7901 gastric cancer cell was weakened but oxaliplatin resistant significantly enhanced;(3)Compared with the parental human gastric cancer cells SGC-7901,metastasisand invasion ability of oxaliplatin resistant human SGC-7901 gastric cancer cell was significantly enhanced;(4)Compared with the parental human gastric cancer cells SGC-7901,the expression of epithelial associated protein(E-cadherin)in oxaliplatin resistant human SGC-7901 gastric cancer cell was decreased and the interstitial associated protein(vimentin)was increased.Conclusions: Although SGC-7901 oxaliplatin resistant gastric cancer cells reduced proliferation,but it has stronger resistance ability and metastasis and invasion ability.At the same time,it has the characteristics of molecular biology related with EMT. |
