Objective: To investigate the effects of ceramic alloys on the expression of oral mucosa’s upstream apoptotic signal molecules.Methods:(1)Five kinds of ceramic alloys(the experimental group),as well as polypropylene(the negative control group)were case to disk type specimens,whose diameter was 5 millimeters while the thickness was 0.5 millimeters.After polished,cleaned and high pressure sterilized,these specimens were fixed on the oral mucosa of golden hamsters.(2)In the fourteenth and twenty-eighth days after suturing the specimens,cut the cheek pouch mucosa contacting with the specimens.Then,we use Real-time qPCR to test the mRNA’s expression of Bcl-2、Bax、Fas、FasL and Cyt-c in the cheek pouch mucosa of golden hamsters.(3)In the fourteenth and twenty-eighth days after suturing the specimens,cut the cheek pouch mucosa contacting with the specimens.Then,we use Western Blot to test the protein’s expression of Bcl-2、Bax、Fas、FasL and Cyt-c in the cheek pouch mucosa of golden hamsters.Results:(1)The results of Real-time qPCR showed that,the mRNA’s expression of Bcl-2 was downregulated in Ni-Cr groups at both the fourteenth and the twenty-eighth days when compared to that of the negative control group at the same time;While the m RNA’s expression of Bcl-2 were upregulated in Co-Cr group at the twenty-eighth day and CP-Ti、Pd-based and Au-Pt groups at the fourteenth day.The mRNA’s expression of Bax was upregulated in Ni-Cr group at both the fourteenth and the twenty-eighth days when compared to that of the negative control group at the same time;While the mRNA’s expression of Bax were upregulated in Co-Cr group at the twenty-eighth day and Pd-based group at the fourteenth day;The mRNA’s expression of Bax were downregulated in Au-Pt group at the twenty-eighth day.The mRNA’s expression of Fas was upregulated in both Ni-Cr and Pd-based groups at both the fourteenth and the twenty-eighth days when compared to that of the negative control group at the same time;While Co-Cr and Au-Pt groups only at the fourteenth day,andCP-Ti group only at the twenty-eighth day.The mRNA’s expression of FasL was upregulated in Ni-Cr 、 Co-Cr and Pd-based groups at both the fourteenth and the twenty-eighth days when compared to that of the negative control group at the same time;While Au-Pt group only at the fourteenth day,and CP-Ti group only at the twenty-eighth day.The mRNA’s expression of Cyt-c was upregulated in Ni-Cr and Pd-based groups at both the fourteenth and the twenty-eighth days when compared to that of the negative control group at the same time;While Co-Cr group only at the fourteenth day.(2)The results of Western blot revealed that the protein’s expression of Bcl-2 was downregulated in Ni-Cr group at both the fourteenth and the twenty-eighth days when compared to that of the negative control group at the same time;While in Co-Cr group only the fourteenth day and Pd-based group only at the twenty-eighth day.The protein’s expression of Bax was upregulated in Ni-Cr、Co-Cr and Pd-based groups at both the fourteenth and the twenty-eighth days when compared to that of the negative control group at the same time.The protein’s expression of Fas was upregulated in Ni-Cr and Co-Cr groups at both the fourteenth and the twenty-eighth days when compared to that of the negative control group at the same time;While in Pd-based group only at the twenty-eighth days.The protein’s expression of FasL was upregulated in Ni-Cr、Co-Cr and Pd-based groups at both the fourteenth and the twenty-eighth day when compared to that of the negative control group at the same time.The protein’s expression of Cyt-c was upregulated in Ni-Cr、Co-Cr and Pd-based groups at both the fourteenth and the twenty-eighth days when compared to that of the negative control group at the same time.Conclusion:(1)In the oral complex environment,different ceramic alloys can have different effects on the mRNA’s and protein’s expression of upstream apoptotic signal molecules.(2)The Ni-Cr 、 Co-Cr and Pd-based alloys can lead to different degrees of apoptosis,and we supposed that the apoptosis may presumably mainly induced by endogenous signal transduction pathways,and there may be other apoptotic signal transduction pathways involved in.(3)The CP-Ti and Au-Pt alloys may not cause apoptosis,and have good biocompatibility.
