Objective: To investigate the effect of ceramic alloys on apoptosis of oral mucosa and apoptotic signaling molecules.Methods:(1)Five kinds of ceramic alloys were cast for one time to gain disk type specimens. Polypropylene were chosen as negative control. All specimens were polished, cleaned with pure water and sterilized at 121.3?for 20 min. Finally, specimens were fixed on the oral mucosa of golden hamsters.(2) Removed the mucosa contacting with the specimens at 14 th day and 28 th day respectively.Embedded the tissues with paraffin and made paraffin sections. Tunel staining was used to detect the expression of cell apoptosis.(3) The m RNAs' expression of Caspase-3?Caspase-8?Caspase-9 in golden hamsters' oral mucosa which were removed at 14 th and 28 th day after fixing the specimens were detected by Real-time q PCR.(4) The proteins' expression of Caspase-3?Caspase-8?Caspase-9 in golden hamsters' oral mucosa which were removed at 14 th and 28 th day after fixing the specimens were detected by Western Blot.Results:(1) The result of Tunel staining reveal that the apoptosis rate of nickel-chromium ?cobalt chromium and palladium based alloys are higher than the negative control.(2) The results of Real-time q PCR revealed that the expression of Caspase-3 of nickel-chromium ? cobalt chromium and palladium based alloys upregulated at both 14 th and 28 th days,While that of CPTi upregulated at only 14 th day. The expression of Caspase-8 of nickel-chromium and palladium based alloys upregulated at both 14 th and 28 th days,while that of cobalt chromium upregulated at only 28 th day. The expression of Caspase-9 of cobalt chromium and palladium based alloys at 14 th day as well as that of nickel-chromium and palladium based alloys at 28 th day upregulated.(3) The results of Western blot revealed that the expression of Caspase-3?8?9 of nickel-chromium ?cobalt chromium?CPTi and palladium based alloys upregulated at 14 th day, while the expressions of only Caspase-8 ? 9 of gold-platinum alloys upregulated. At the 28 th day,the expression of Caspase-3 of nickel-chromium ?cobalt chromium ?CPTi and palladium based alloys upregulated and the expression of Caspase-8 ofnickel-chromium?palladium based alloy upregulated. For the expression of Caspase-9, all kinds of alloys except cobalt chromium alloy upregulated.Conclusion:(1)The nickel-chromium ?cobalt chromium and palladium based alloys may induce apoptosis of golden hamster's oral mucosa.(2)The five kinds of ceramic alloys can induce different extent expressions of apoptotic signaling molecules Caspase-3?8?9 at m RNA level or protein level in golden hamster's oral mucosa.
