A Preliminary Study On The Effect Of Folate On Autophagy Protein P62 In SiHa And HeLa Cells

Objective:To investigate the effect of folate on the expression of autophagy related protein p62 in cervical cancer Si Ha cells and He La cells.Methods:The cervical cancer Si Ha cells and He La cells were treated with two levels of folate level(normal control group,1.0?g/ml)and low dose of folate level(low dose of folate group,0.1?g/ml).Cells were collected at 24 hours,48 hours and 72 hours respectively.Real time fluorescent quantitative PCR assay and Western blot assay were used to detect the expression of p62,a key protein of autophagy,in m RNA and protein level.Results:1.In the cervical cancer Si Ha cells,compared with the normal control group,the expression levels of p62 were decreased at 48 hours and at the same time for 72 hours in the low dose of folate group(P<0.05).The expression level decreased only 72 hours at m RNA level(P<0.05).2.In the cervical cancer He La cells,compared with the normal control group,in the low dose of folate group the expression level of p62 decreased at 48 hours was decreased only at 48 hours in protein level and m RNA level(P<0.05).Conclusion:1.Low dose of folate decreased the expression of autophagy related protein p62 in cervical cancer Si Ha cells,suggesting that low dose of folate could induce autophagy in cervical cancer cells.2.Low dose of folate decreased the expression of autophagy related protein p62 in cervical cancer He La cells,also suggesting that low dose of folate could induce autophagy in cervical cancer cells.
