Effect Of MiRNA-195 On The Expression Of CCND1 In Glioma U251 Cells

Objective:To study the expression of CCND1 in glioma drug-resistant cells and explore the relationship between CCND1 and glioma drug-resistant cells,so as to provide a basis for elucidating the application value of CCND1 in glioma chemotherapeutics.Methods:Through the liposome Lipofectamin 2000 MiRNA-195 mimics,MiRNA-195 inhibitor was transfected into U251 glioma cells,the content of MiRNA-195 Real-time before and after PCR was detected to ensure transfection efficiency;through the expression of Real-time before and after PCR transfected CCND1mRNA;the expression level of CCND1 Western Blotting detection in U251 glioma cells;SPSS13.0 software was used to analyze the data,quantitative data is described by (?)� s,data comparison between groups using ANOVA.The comparison between the 22 groups using LSD-t test,P<0.05 difference was statistically significant.Results:1.The expression of CCND1 mRNA in U251 glioma cells transfected with MiRNA-195 mimics was higher than that of U251 glioma cells transfected with MiRNA-195inhibitor;2.The expression of CCND1 protein in U251 glioma cells transfected with MiRNA-195 mimics was higher than that of U251 glioma cells transfected with MiRNA-195 inhibitor.Conclusion:1.MiRNA-195 has an effect on the expression of U251 in glioma cell line CCND1;2.Down regulation of CCND1 expression in glioma U251 cells by overexpression ofMiRNA-195.
