| ObjectiveDynamicly observed resistting gliomas growth and the expression of miRNA-21 and miRNA-221 in Siweixiaoliuyin group and the control groups for explorring the treatment of glioma.Investigated the upstream and downstream target genes about the therapeutic effects of Siweixiaoliuyin by affecting miRNA-21,miRNA-221.At the same time,studied and analized the clinical effect and mechanism of Siweixiaoliuyin treatment with glioma.Methods1.The basic research is about the growth of tumor and miRNA-21's,miRNA-221 ' s expression of Siweixiaoliuyin group and other groups in nude mice model.(1)The single cell suspension of U87 were injected subcutaneously in each mouse flank.(2)The nude mice were quasi-randomized divided into group A(0.9%N.S.),group B(Siweixiaoliuyin),group C(TMZ),and group D(Siweixiaoliuyin+TMZ)randomly for intragastric administration last 28 days.(3)We detected the expression of miRNA-21 and miRNA-221 by qPCR.2.The clinical part is evaluation of Siweixiaoliuyin in clinical effects of patients with glioma,and exploring correlations between it and serum of miRNA-21 and miRNA-221.(1)60 patients whom meet the requirements from the Department of Neurosurgery were chose.With the randomization and control principle,they wre divided into 2 group(30 cases of Siweixiaoliuyin treating group and 30 cases of control group).(2)Before and after different treatments,the clinical symptoms of patients should be observed and marked.Evaluated the quality of survivals by Karnofsky performance score.Monitored blood routine,liver function,kidney function,lymphocytes.Detected miRNA-21,miRNA-221 before and after treatment by qPCR and other related indicators.Results1.In animal experiment,the expression of miRNA-21 and miRNA-221 in Siweixiaoyin group were lower than the negative group(p=0.043?0.049?0.037<0.05;0.048?0.016?0.036<0.05).The combination' s expression of miRNA-21 was lower than another three groups(p=0.005?0.000?0.000<0.05;0.002?0.001?0.000<0.05;0.025?0.005?0.036<0.05),while miRNA-221 as same as miRNA-221(p=0.010?0.001?0.011<0.05;0.005?0.001?0.002<0.05;0.033?0.049?0.024<0.05).2.The expression of serum in clinical research treatment group is lower than the negative(p<0.05).3.In clinical research,the KPS in treatment group(after 28 days treatment)was higher than the control one(P=0.000<0.05),and its integral peak of TCM syndrome comparing with the other group was decreased(P=0.026<0.05).The KPS in Siweixiaoliuyin group has significantly statistical differences between before treatment and after 7 days treatment(p=0.010<0.05),as same as between before treatment and 28 days(p=0.000<0.05).4.Self controlled in the same individuals,the treatment group' s proport-ion of CD3+CD8+ were falling(p=0.001?0.000?0.003<0.05),howover,the negative group were rising(p<0.05).Especially,after treatment,Siweixiaoliuyin group' s proportion is lower than the negative(p=0.006<0.05).Self controlled in the same individuals,the treatment group' s proport-ion of CD4+/CD8+ were rising(p=0.001?0.000?0.000<0.05).The negative group were falling(p<0.05).And after treatment,Siweixiaoliuyin group is higher than the negative(p=0.047<0.05).Comparison between groups,surprisingly,two groups had no statistical differences of CD3+CD4+(p>0.05).5.When the research finished,two groups of RBC,Hb,AST,ALT,Cr and BUN had no statistical differences(p>0.05).Conclusion1.Siweixiaoliuyin could inhibit nude mice glioma tumor growth by regulating the exoression of miRNA-21 and miRNA-221.2.Siweixiaoliuyin combined with TMZ might inhibit the glioma tumor growth rate,and reduce the expression of tumor' s miRNA-21 and miRNA-221,for strengthen the glioma sensitivity to chemotherapy drugs.3.Siweixiaoliuyin traement would be a new therapy in glioma.It might improve the patients' immunity,syndrome in TCM and the quality of lives.4.Using Siweixiaoliuyin did not appear harmful response about liver and kidney function.It would reduce the toxicity of chemotherapy. |
