Objective To study the expression of Survivin,Caspase-3 and Ki-67 in glioma and its significance. This study will provide some experimental evidences for the development mechanism of glioma and provide a new target for the gene diagnosis and therapy .Methods Immunohistochemical technique(ElivisionTM plus)was used to detect the expression of Survivin,Caspase-3 and Ki-67 protein in 68 cases of primary glioma and 15 cases of recurrent glioma and 10 cases of normal brain tissue. In situ hybridization technique was used to detect the expression of Survivin and Caspase-3 mRNA in glioma and normal brain tissues. TUNEL method was used to detect cell apoptosis about 68 cases of primary glioma.Results 1,①There was a significant difference between normal tissue and glioma about the positive rates of Survivin and Ki-67 protein,the same as in different grade glioma. The positive expression of Survivin protein in grade III,IV glioma were obviously higher than grade I,II glioma,the same as the Ki-67. There was a significant difference between primary and recurrent glioma about the strong positive rates of Survivin protein,the same as the Ki-67 protein.②There was not significant difference between normal tissue and glioma about the positive rates of Caspase-3 protein,the same as in different grade glioma. There was not significant difference between primary and recurrent glioma about the strong positive rates of Caspase-3 protein.③Correlation analysis according to Spearman revealed a significant positive correlation between the expression of Survivin protein and Ki-67 protein in glioma,a significant negative correlation between the expression of Survivin protein and Caspase-3 protein in glioma and a significant negative correlation between the expression of Caspase-3 protein and Ki-67 protein in glioma.2,①There was a significant difference between normal tissue and primary glioma about the positive rates of Survivin mRNA. The positive expression of Survivin mRNA in grade III,IV glioma was obviously higher than grade I,II glioma.②There was not significant difference between normal tissue and primary glioma about the positive rates of Caspase-3 mRNA.③Correlation analysis according to Spearman revealed a significant negative correlation between the expression of Survivin mRNA and Caspase-3 mRNA in glioma.3,The mean AI of the 68 cases of primary glioma was 16.59%(SD,6.19%; range, 7–29%). AI in the groups of positive expression of Survivin and Ki-67 are significantly lower than their negative groups. AI in the group of positive expression of Caspsae-3 was significantly higher than the negative group.Conclusions1,The strong expression of Survivin and Ki-67 can be regarded as one of the high risk factors about the recurrence of glioma.2,The high expression of Survivin and Ki-67 can be regarded as the signal about the dysdifferentiation of glioma.3,There was not significant difference between normal tissue and primary glioma about the expression of Caspase-3. So do in difference grade glioma.4,Survivin probably inhibits Caspase-3 expression directly or indirectly,which results in the occurring and progression of glioma. Survivin and Ki-67 play a coordination or accommodation part in the occurring and progression in glioma. The occurring and progression of glioma are associated with the reduction of apoptosis and the reinforcement of proliferation.5,Mean AI in the group of positive expression of Survivin is lower than the negative group. Mean AI in the group of positive expression of Caspase-3 is higher than the negative group.6,Survivin mainly acts on the cross point of apoptosis way,and only exists in tumor tissue,so it is one of the most ideal targets for cell apoptosis therapy . Survivin will become a new direction for gene diagnosis and targeted therapy of glioma.
