The identification characteristics,chemical composition and pharmacological action of Red coral were studied in this paper,which provide the basis for future research and application of Red coral.First of all,the taxonomy,resources distribution,and formation of skeletons,morphological characteristics,and medicinal value in traditional medicine,chemical components and pharmacological effects of Red coral were reviewed in this paper.Traits identification showed that skeleton of Red coral take the dendritic shape or short rods.They are different in size and are vertical lines on the surface.In terms of microstructure,to observe the Red coral powder under the optical microscope,visible with angular particles,these particles showed translucent,dark edges.In addition,can also be seen the needle or flake crystal in Red coral powder.A small amount of coral tentacles are visibled in the powder.The main features observed by scanning electron microscopy(SEM)are that the Red coral surface is composed of a plurality of raised structures formed by the ring-shaped stack.Its cross-section shows a number of concave structures,pointing to the center of the spine to form this concave structure.The tightly arranged spur is observed on the longitudinal aspect of the Red coral.Physical and chemical identification,the color of Red coral has not changed after the organic reagent soaked.And has a bubble phenomenon after drop of hydrochloric acid on the Red coral.Chemical component,(1).In the aspect of inorganic elements: rich macro elements and essential trace elements contained in Red coral followed by Ca(62200 mg/kg),Mg(13310 mg/kg),Sr(2373 mg/kg),Na(1494 mg/kg),Fe(1321 mg/kg),K(198 mg/kg),Zn(25.59 mg/kg),Ba(13.65 mg/kg),Mn(1.839 mg/kg),Cu(1.465 mg/kg),Cr(0.6567 mg/kg).(2).In the aspect of organic ingredients: there were 16 kinds of compounds were obtained from liposoluble component of Red coral that include alkane,lipid,alcohols,squalane,and other organic compounds,which contained 12 substance reported for the first time.Among them,the relative content of dibutyl phthalate is 70.239 %,1,2-phthalate(2-methyl propyl)ester 7.168 %,single dimethyl phthalate cyclohexyl 6.232 %.Gradient extraction method was used to extract organic compounds of Red coral.The chemical constituents were isolated and purified by silica gel,Sephadex LH-20 and HPLC chromatography,from powder of Red coral.The structures of the isolated compounds were elucidated by spectroscopic analysis and physicochemical properties.Five compounds were isolated and identified,they were diisobutyl phthalate(1),dibutyl phthalate(2),carbazole(3),p-hydroxy benzoic acid(4),6(5h)-Phenanthridinone(5);They were all for the first time found from Red coral.The study of pharmacological activity,(1).Pharmacological effects for activating blood circulation,using blood stasis model rats,to study the effect and mechanism of Red coral on hemorheology in model rats by using aspirin as positive control.Studies have shown that when the rat by “rage,cold evil” after the invasion can cause vascular wall damage,liver occasionally point necrosis,oral administration of Red coral can reduce vascular damage and liver necrosis and other lesions;Red coral can reduce the whole blood viscosity and plasma viscosity value at different shear rates,the difference was statistically significant(P<0.05);Its mechanism of action is associated with regulation of platelet aggregation.The results showed that Red coral could decrease the plasma TXB2 content in the blood stasis model rats,increase the level of 6-keto-PGF1?,the difference was statistically significant(P<0.05).At the same time,Red coral could decrease the levels of plasma PF4,?-TG and ET-1,increase the level of NOS,the difference was statistically significant(P <0.05).(2).Aspect of the promote fracture healing,the radial fracture model of rats with Red coral as the research object,through the X-ray and HE staining more intuitive evaluate the change of the fracture,and combining with index of serum ALP,Ca and P content changes.The X-ray image shows,in 10 days after the treatment,the rats fracture line edge blur of Red coral groups,In this region of a recent fracture,the 20 days after,callus is seen forming at the broken ends of bony trabeculae,increased of the bone mineral density and edge closer to disappear.In the 30 th day when fractures healed completely.A fracture position HE dyeing can be seen,the fibre organization increased of rats in Red coral dose group,and the porosis faster compared with other groups of rats.The high dose of Red coral groups rat has obvious promotes healing fracture better and faster than other groups.The levels of serum calcium(Ca)and phosphorus(P)in high dose group were significantly higher than those in model group,and the content of alkaline phosphatase(ALP)change occurred from increase to decrease,there is significantly differences(P<0.05).This article provided a basis for the identification of Red coral herbs.The 16 compounds were analyzed by GC-MS from the petroleum ether extract.And 5 compounds were obtained from Red coral extracts.Experiments show that Red coral has the effect of promoting fracture healing,the mechanism may be related to the regulation of Ca,P,ALP phosphatase and affect the body's blood rheology.
