| 【Objective】To observe the histopathological features of refractory nasal sinusitis with nasal polyps and common nasal sinusitis with nasal polyps,To explore the histopathological differences between the two groups,and investigate the pathological basis of refractory nasal sinusitis with nasal polyps,and to provide a reference for further study of molecular mechanism and postoperative follow-up and medication.【Methods】1.All the cases are taken from the Department of Otolaryngology of the Affiliated Hospital of Qiingdao University Between September 2011 to September 2015 which are choosed by the inclusion and exclusion criteria,and collect cases’ data that meet the admission criteria.2.The cases were divided into 2 groups:The refractory group and The control group,there were 30 cases in each group.To collect the pathological wax block according to the admission number,each wax block was marked with a number of pathological marks.All pathologic wax blocks were disrupted after the pathological numbers of the refractory group and the control group were recorded separately,then randomly select wax block to reordering.3.Dewaxing,sectioning and staining the pathological wax block,processed for histological studies.Research method:(1)The number of inflammatory cells(include Eosinophils,Neutrophils,Plasmacyte,Lymphocyte)were counted under a light microscope with Conventional HE staining,(2)Using immunohistochemical EnVision method staining the pathological sectionsCounting the number of CD34 labeled positive microvessels in the refractory and control groups.All the data were statistically analyzed.4.Pathological sections treated by HE staining and immunohistochemistry were determined according to the experimental method by the pathologists who did not know the grouping and sorting.5.All calculations were performed with SPSS 17.0 using t test,and all data was expressed as(x±s).The level of significance was considered to be P <0.05.【Results】1.The number of Eosinophils,Neutrophils,Plasmacyte,Lymphocytes and Microvessel density(MVD)in the group of the refractory group and the control group were compared by independent samples t test,t Values were5.009、2.308、2.414、2.026、4.025,P<0.05,a statistically significant difference.2.The number of Eosinophil and Microvessel density have positive correlation(r=0.636、0.558,P<0.01)in two groups and there was a negative correlation between lymphocyte and microvessel density(r=-0.509、-0.433,P<0.05).【Conclusion】1.The number of eosinophils and the number of CD34 positive microvessels were significantly increased in patients with refractory rhinosinusitis and nasal polyps2.There was a positive correlation between the degree of eosinophil infiltration and microvessel density.3.It is possible to determine the prognosis of nasal polyps according to the number of eosinophils and microvessel density in clinical. |
