Research On Mouse BMSCs Transplantation For Improving Ovarian Reservation In A Rat Model Of Premature Ovarian Failure

Objective To investigate the effect of transforming growth factor ? receptor ?(TGF-? R?)and transforming growth factor ? receptor ?(TGF-? R?)on the transplantation of bone marrow mesenchymal stem cells(BMSCs)which were studied in the treatment of premature ovarian failure(POF)rats.And then the research provides a new scientific basis for the use of BMSCs on the treatment of POF.Methods Female SD rats were divided into groups(control group,model group,natural recovery group,and BMSCs transplantation treatment group),then POF models were established by intraperitoneal injection of cyclophosphamide.GFP transgenic SD rats were selected for BMSCs,and analyzed by immunofluorescence technique and flow cytometry,The BMSCs transplantation treatment group was treated by BMSCs through caudal vein injection.The estrous cycles of all groups were observed.The serum of each group were detected by ELISA method to test E2 and FSH levels in venous blood.Ovarian tissue were detected the expression of TGF-?R?and tgf-beta 1 R,among groups by HE staining,immunohistochemistry,Western blot(Western blot),fluorescent quantitative PCR technology(real-time PCR).The results were statistically analyzed by SPSS 22.0 software,and differences were regarded as statistically significant for two-tail values of P<0.05.Results 1.The expression of TGF-? R ?m RNA significantly up regulated in the control group and the BMSCs transplantation treatment group,differed from expressions of the model group and the natural recovery group(P < 0.05),while the expression of TGF-? R ?m RNA had no statistical significance in each group(P > 0.05).2.The expression of TGF-? R ?protein in the control group and the BMSCstransplantation treatment group were up regulated which had statistically significant(P < 0.05).3.There were positive expression of TGF-? R ?in the ovarian tissues of control group and BMSCs transplantation group.4.The E2 level detected by ELISA in the BMSCs transplantation treatment group was distinctly higher than it in the model group and the natural recovery group(P < 0.05),while the FSH level of the BMSCs transplantation treatment group was lower than it in the model group and the natural recovery group(P < 0.05).5.In the BMSCs translation treatment group,there were all levels of follicle cells,while there were few primordial follicles in the natural recovery group,and no follicle in the model group.6.In the BMSCs transplantation group,the estrous cycle was gradually resumed.In the natural recovery group,one or two of them appeared normal cycle.The estrous cycle of the model group was stationary.7.CD90 and CD105 were expressed on the BMSCs membranes.Flow cytometry showed that the fluorescence intensity of CD90 and CD105 was in accord with the experiment.Conclusions 1.Mouse BMSCs transplantation can improve the ovarian reserve function of POF animal model.2.TGF-?R ?has a great role in repair of premature ovarian failure after bone marrow mesenchymal stem cells transplantation.3.The distribution of scattered fluorescent cells was observed in the ovarian vessels and tissues,which affirms the role of BMSCs homing.4.The positive rate of CD90 markers detected by flow cytometry is 98.9%,which indicates that bone marrow mesenchymal stem cell transplantation may have a wide application prospect on the treatment of premature ovarian failure.
