Objective To observe the effect of capsaicin on the symptoms,nasal mucosa morphology and substance P expression and its effect in allergic rhinitis rats,and to explore the mechanism of capsaicin in the prevention and treatment of allergic rhinitis.Methods 1.A total of 30 healthy SD male rats were randomly divided into normal control group,AR group and capsaicin intervention group(n=10),pay attention to their scratching nose,foraging,rhinorrhea,sneezing and other acts;2.To collect the nasal mucosa of rats,give HE staining,toluidine blue staining and immunohistochemical staining to them and then observe the Eosinophils infiltration degree,mast cell and the content of substance P;3.Using ELISA method to examine the serum Ig E、ECP、OVA-s Ig E levels in each group.Results In the AR group and the capsaicin intervention group,SD rats showed sneezing,runny nose,irritability,sore nose and tickling in the 30 min after the last nasal challenge.The symptom score was> 5,the two groups were successful modeling,the normal control group did not appear sneezing and other AR performance.The difference was significant(P <0.05).The scores of SD rats in the AR group were(6.9 ± 0.99),and the average score of the SD rats in the capsaicin treatment group was(7.1 ± 0.73).After 15 days of capsaicin treatment,the average score of SD rats in SD group was(6.40 ± 1.42),the average score ofSD rats in capsaicin treatment group was(4.10 ± 1.66)and 0.90 ± 0.31 in normal control group.There was no significant difference between the two groups before and after treatment(t = 1.627,P = 0.138> 0.05).The symptom score was lower in the capsaicin treatment group(t = 5.809,P <0.001).2.HE staining see normal control group nasal mucosa tissue structure is normal,no inflammatory cell infiltration.The mucosa’continuous were interrupted,cilia were lodged or even falled off,eosinophils,mast cells were infiltrated obviously in AR group.Capsaicin treatment group,mucosal epithelial injury was significantly improved,hypertrophy of goblet cells decreased,glands and dilated ducts similar to the normal control group,in the lamina propria,inflammatory cells were significantly reduced,but not completely eliminated.The number of mast cells in the AR group was higher than that in the normal control group(P<0.05).The results were statistically significant.Capsaicin treatment group mast cell count was significantly lower than the AR group,P<0.05,the results were statistically significant.The content of substance P in nasal mucosa of AR group was significantly higher than that of normal group(P<0.05).The content of substance P in nasal mucosa of capsaicin treatment group was lower than that of AR group.The difference was statistically significant.The results of ELISA showed that serum Ig E,ECP and OVA-s Ig E decreased in the capsaicin treatment group compared with AR group(P<0.05).Conclusions 1.Capsaicin nasal drops can reduce the symptoms of allergic rhinitis in SD rats.2.Capsaicin nasal therapy can make SD rats nasal mucosal tissue mast cells,eosinophils decreased,vasodilatation reduction.3.Capsaicin nasal therapy can reduce the expression of substance P in nasal mucosa of SD rats.4.Capsaicin nasal drops can reduce serum Ig E,ECP,OVA-s Ig E levels in SD rats,and thus inhibit the development of allergic rhinitis.
