| Objective: To evaluate the therapeutic effectand mechanism after the treatment of Yangying Huoxue decoction on hepatic fibrosis rats,by investigating its difference on related Serum hepatic index and liver fibrosis degree?serum ?-SMAandTGF-?1 in each group.Methods: A total of 72 specific pathogen free(SPF)Wister rats(male,200±20g)were randomized into 6 groups as normal group,model group,colchicine treatment group and Yangying Huoxue decoction high dosage,medium dosage and low dosage treatment groups.The HF rat model was mainly established by 40% CCL4 mixed with paraffin oil and porcine serum.Giving corresponding drugs to each group for 8 weeks after 4 weeks of successful modeling.Detect the related biochemical indicates of rat serum,at the same time,observe and compare the pathological results of liver tissue after HE straining for each group,liver tissue homogenate was prepared to detect the content of TGF-1 and ?-SMA by ELISA.Results: The Yangying Huoxue decoction treatment group can effectively lower the ALT?AST and ?-GT content in HF rats,rise the contents of ALB.Compared with model group,after the treatment with high,medium and low-dose of Yangying Huoxue decoction,there were different degrees of reduction in liver hyperplasia of fibrous tissue,edema and steatosis.the TGF-?1 and ?-SMA were significantly lower than model group,shows statistically significant(P<0.05 or P<0.01).Compared with colchicine treatment group,the concentration of?-SMA and TGF-?1 were significantly decreased in the Yangying Huoxue decoction high-dose treatment group,shows statistically significant(P<0.05).Although the absolute value of the Yangying Huoxue decoction medium-dose treatment group was decreased,it wasnot statistically significant(P>0.05).The concentration of the Yangying Huoxue decoction low-dose treatment group was higher and the difference was statistically significant(P<0.05).In the comparison result of liver fibrosis grading,the fibrosis of each treatment group was lighter than that of the model group,and the difference was statistically significant(P<0.05);Compared with colchicine treatment group,the effect of Yangying Huoxue decoction high-dose treatment group was better and statistically significant(P<0.05).the same effect as medium-dose treatment group;and the low-dose treatment group was less effective than the control group,which was statistically significant(P<0.05).Conclusion: Yangying Huoxue decoction can obviously alleviate rat hepatic fibrosis grade,improve the related serum hepatic index,lower the concentration of ?-SMA,which mayrelated with reducing serum TGF-?1 levels. |
