Dextran Sulfate Inhibits Cell Migration And Invasion Through LOX In Gastric Cancer | Posted on:2018-08-13 | Degree:Master | Type:Thesis | Country:China | Candidate:X F Wang | Full Text:PDF | GTID:2334330536969575 | Subject:Pathology and pathophysiology | Abstract/Summary: | PDF Full Text Request | Objective To investigate the inhibitory effect of dextran sulfate(DS)on human gastric cancer cells(BGC-823)under hypoxic conditions and explore whether lox play a role in inhibition of the invasion and migration of human gastric cancer cells(BGC-823)by DS.Methods 1.BGC-823 cells were cultured under normal oxygen condition.The proliferation of human gastric cancer cells(BGC-823)was detected by CCK8 assay after 24 hours exprosure of DS and BAPN(LOX active inhibitors)alone or in combination.2.Apoptosis of BGC-823 cells was detected by flow cytometry(DS).3.Under hypoxia,we evaluate whether there is a difference of invasion and migration of BGC-823 cells by invasion and migration assays under the condition of exprosure of DS and BAPN(LOX active inhibitors)alone or in combination.4.BGC-823 cells in the experimental and control groups were administered DS and PBS,respectively,and exposed to hypoxic conditions for 2h,8h,12 h and 24 h.Immunocytochemistry,immunofluorescence,RT-PCR and western blot analyses were used to evaluate LOX and TGF-? expression levels.5.The expression of LOX,TGF-? and E-cadherin were detected by western blot after BGC-823 cells exposed to DS and BAPN alone or in combination.Results 1.24 hour exposure of DS and BAPN inhibited the proliferation of BGC-823 cell lines in a concentration dependent manner deceted by CCK8.The combination DS and BAPN exprosure achieved the most significant inhibition of proliferation of BGC-823 cell lines.2.24 hour exposure of DS incresed the apoptosis of BGC-823 cell lines also in a concentration dependent manner deceted by Flow cytometry.3.DS and BAPN could inhibit the invasion and migration of BGC-823 cells under hypoxic condition by Transwell test(P <0.05);The combination DS and BAPN exprosure achieved the most significant inhibition of invasion and migration of BGC-823 cell lines(P <0.05).4.The expression of LOX and TGF-? was in a time dependent manner.DS significantly decreased LOX and TGF-? expression at different exprosure time with significant difference by Immunocytochemistry and immunofluorescence.5.Compared with the control group,DS could significantly reduce the expression of LOX mRNA and TGF-?mRNA at different exprosure time with significant differences by RT-PCR;DS can increase significantly the expression level of Smad4 mRNA at different exprosure time.6.DS could significantly reduce the expression level of LOX and TGF-? at different exprosure time by Western blotting.Compared with the control group,DS increased the expression level of Smad4 and E-cadherin at different exprosure time.Combined DS and BAPN 12 h suppressed significantly the expression of LOX and TGF-?,while increased the expression of E-caherin significantly.Conclusion1.DS affects on the proliferation and apoptosis of human gastric cancer cells in a concentration-dependent manner.2.DS can inhibit the invasion and migration of human gastric cancer cells.The inhibition of DS combined with BAPN is more significantly.3.DS can reduce the expression of LOX and TGF-? in human gastric cancer cells and increase the expression of Smad4 and E-cadherin.The effect of DS combined with BAPN is more significantly.LOX expression was positively correlated with TGF-? expression.LOX may play an important role in inhibition of the invasion and migration of human gastric cancer cells(BGC-823). | Keywords/Search Tags: | Dextran sulfate, Human gastric cancer cells, Lysyl oxidase, Invasion, Migration | PDF Full Text Request | Related items |
