Objective To study the effects of Lysyl Oxidase (LOX) on Gastric cancer cell SGC-7901's adherence and migration by exposing them to different concentration of the LOX inhibitor,β- aminopropionitrile (BAPN) and specific anti-LOX antibody. To evaluate the effect of LOX on human umbilical vein endothelial cell (HUVEC) angiogenesis through treating the cells with BAPN.Methods Gastric cancer cells SGC-7901 and HUVEC were cultured in vitro. After exposing gastric cancer cells SGC-7901 to different concentration of BAPN (0mM, 0.1mM, 0.2mM and 0.3mM) and anti-LOX antibody (0μg/ml, 5.0μg/ml, 10.0μg/ml and 15.0μg/ml), the abilities of adhesion between cell-cell and cell-ECM were detected by homogeneity and heterogeneous adhesion experiments, and the ability of migration was assessed by wound-healing assay. When exposing the HUVEC to different concentration of BAPN (0mM, 0.1mM, 0.2mM, and 0.4mM), the abilities of HUVEC angiogenesis induced by VEGF, bFGF and PMA were checked using fibrin gel in vitro angiogenesis assay kit.Results 1. After treated by 0.1mM,0.2mM,0.3mM BAPN,the homogeneity adhesion of gastric cancer cells SGC-7901 was increased. Added 5.0μg/ml,10.0μg/ml,15.0μg/ml anti-LOX,the homogeneity adhesion of gastric cancer cells SGC-7901 was enhanced.2. After treated by 0.1mM,0.2mM,0.3mM BAPN,the heterogeneous adhesion of gastric cancer cells SGC-7901 was attenuated. And added 5.0μg/ml,10.0μg/ml,15.0μg/ml anti-LOX,the heterogeneous adhesion of gastric cancer cells SGC-7901 was down regulated.3. Treated by 0.1mM,0.2mM,0.3mM BAPN,the migration of gastric cancer cells SGC-7901 was descended. Meanwhile, added 5.0μg/ml,10.0μg/ml,15.0μg/ml anti-LOX,the invasion of gastric cancer cells SGC-7901 was decreased.. 4. The results of fibrin gel in vitro angiogenesis assay showed angiogenesis of HUVEC were induced by VEGF, bFGF, PMA, and these inductions were inhibited by BAPN.Conclusion BAPN and anti-LOX can enhance homogeneity adhesion,attenuate heterogeneous adhesion, and weak the migration of gastric cancer cells SGC-7901. These suggest that LOX promote the metastasis of cancer cell through enhancing migration,increasing heterogeneous adhesion and decreasing homogeneity adhesion. The angiogenesis of HUVEC induced by VEGF, bFGF, PMA was inhibited by BAPN. The results could reveal that LOX may promote angiogenesis induced by angiogenesis factor.
