Objective:To observe the effect of Guizhi Fuling pills on inflammatory cytokines and adhesion related immune molecules in different stages of PID ratsand to provide theoretical basis for the selection of clinical medication time and the choice of medication methods.Methods:The PID model was established by SPF class female non pregnant SD rats,The mechanical injury,endometrial infection and bacterial infection were adopted,One hundred and ninety-eight rats were randomly divided into eight groups:sham operation group and control group;model group,treatment group(GFW high,medium and low dose group),control(gold group Gangteng group and metronidazole group).Drug intervention resspectively seven days,Fourteen days,Twenty one days,were randomly selected in rats at different times,weighing,take the blood of rats;removal of uterus and ovary tissue were observed by naked eyes and weighing,respectively the index calculation;different time groups in rat serum proinflammatory cytokine interleukin-8(IL-8)and anti-inflammatory cytokine interleukin-10(IL-10)by used enzyme-linked immunosorbent assay(ELISA method)to determine the pathological changes;The pathological changes of uterus tissue in different time were observed under light microscope;uterine tissue and immune molecules of matrix metalloproteinase-9(MMP-9)and vascular endothelial growth factor(VEGF)expression by immunohistochemical method(S-P method)to detect.Results:1 The model rats appeared similar to cold dampness stagnation syndrome,comparedwith blank group,sham operation group,different drugs intervention,the rats of model group were improved in general different time.2 model group and blank group,sham operation group,weight gain,after drug intervention groups in different time weight decreased,the difference was statistically significant(P<0.05),with twenty-first days of Guizhi Fuling pill high dose groupsignificantly(P<0.05),different time in each group was not statistically significant(P>0.05).3 model group the uterus index and ovarian index were decreased significantly compared with control group and sham operation group(P<0.05);compared with the model group,after the intervention of the drugs in seventh,Fourteen,twenty-one days,increased the uterus index and ovarian index in different extent,the changes were statistically significant(P<0.05).There was no significant difference between the groups(P>0.05).4 the level of IL-8 and the level of IL-10 decreased in the model group,and the relationship between them was negatively correlated.There was statistical significance compared with control group and sham operation group(P<0.05),with the extension of time,the intervention of different drugs,the IL-8 decreased,IL-10 increased in different time in each group eventually recovered close to the normal level,especially Guizhi Fuling bolus dose group was more obvious(P<0.05).5 The endometrium of rats in the model group had abnormal changes at different time periods.The main performance is:the majority of epithelial cells necrosis in seventh days,infiltrating lymphocytes,monocytes and other inflammatory cells,endometrial have obvious vascular congestion expansion;fourteenth days when the endometrial structure is unclear,disorganized,connective tissue hyperplasia,uterine secretion;twenty-first days of muscle fiber texture disorder that part of the gland necrosis and were filled with fibrous connective tissue,uterine cavity showed a large amount of secretions,improve the performance of each group after treatment.6 the expression of VEGF in each model group increased gradually at different time,and there was significant difference between the blank group and sham operation group(P<0.05).After drug intervention,compared with the model group,different time VEGF were lower,but the seventh day dose group,Jingangteng in Guizhi Fuling pills group significantly(P<0.05);dose of Guizhi Fuling pill in Fourteenth days,twenty-one days when the most significant(P<0.01);drug intervention at different time within each group there was no significant difference(P>0.05).Conclusions:1 Guizhi Fuling pill by temperature by promoting blood circulation by removing blood stasis,besides the improvement of different time of general conditions,body weight and uterus,ovarian index such as rats,can also improve the pathological morphology of uterine tissue,the Guizhi Fuling pill high dose group significantly.The therapeutic effect can be achieved by adjusting the disorder of function and metabolism in rats.2 Seventh days PID rat model is still in the acute stage,detoxification method and antibiotic treatment was fourteenth days;the chronic phase,the development trend of Guizhi Fuling pill by temperature by promoting blood circulation and removing blood stasis can effectively regulate the chronic phase of IL-8,IL-10,PID could raise the chronic inflammatory cytokines and reduce the expression of proinflammatory a cell factor for treatment of PID,thereby reducing the possibility of further development or progression of PID.3 Detoxification method in acute exacerbation of PID,can significantly regulate the expression of adhesion related immune molecules,Guizhi Fuling pill by temperature by Huoxue Huayu method can inhibit the expression of PID in chronic phase adhesion related immune molecules,suggesting that it may be to reduce the pelvic tissue by reducing MMP-9,VEGF pelvic adhesion,improve microcirculation,thereby reducing blood vessels the permeability,promote the clearance of damaged tissue,so as to achieve the purpose of treatment of PID.
