Objective To test the radiosensitizer effect of Diallyl disulfide(DADS)on human renal carcinoma 786-0 cells.Methods The cytotoxicity of DADS on 786-0 cells was tested by CCK-8 assay at a concentration of 0,2.5,5,10,20,40 ?mol/L respectively.The clonogenic assay and CCK-8 assay were performed to evaluate the cell colony-forming efficiency and surviving fractions.The cell apoptosis after radiation was analyzed by flow cytometry analysis.Apoptosis paralleled morphological changes were detected with PI fluorescent staining.Bcl-2 and Bax protein expression was measured by Western Blotting.Rsults CCK-8 assay showed that DADS decreased the proliferation of 786-O cells in a dose-dependent manner(F=2403.160,P<0.05).Pretreatment of 5 ?mol/L DADS in 786-0 cells exposed to radiation resulted in decrease in cell viability(F=543.821,P<0.05)and increased radiosensitivity(F=383.136,P<0.05).Additionally,Flow cytometry analysis showed that pretreatment with DADS obviously promoted radiation-induced cell apoptosis(F=764.424,P<0.05),and the results of PI fluorescent staining also showed that combined group had higher level of cell apoptotic.The Western Blotting results indicated that treatment with DADS and irradiation up-regulated the expression of Bax(F=118.350,P<0.05)and down-regulated the expression of Bcl-2(F=396.214,P<0.05)compared with other groups.Comclusion Combination DADS and the radiation exacerbated the activation of apoptosis pathways through up-regulated ration of pro-apoptotic Bax to antiapoptotic Bcl-2.This result suggest that DADS is a potential candidate as radio sensitive agent for renal carcinoma.
