Background: Leukemia is the most commonly malignant tumor in children.Diallyl disulfide is one effective component of garlic, it can induce apoptosis ofdifferent tumor cell lines. C-jun, a member of the AP-1family transcription factorsthat play a key role in normal and abbrent cell growth, moreover, it also involves inapoptosis. It is not clear about the role of c-jun in apoptosis induced by diallyldisulfide in k562cells.Objective: Exploring the role of c-jun in process of apoptosis induced by diallyldisulfide(DADS) in K562cells and elucidating the mechanism of apoptosis inducedby DADS in K562cells. It will supply potential targets for the treatment of leukemia.Furthermore, it establishs partial molecular fundament for applying DADS into theclinical in future.Methods: Through inversion microscope, the morphological change of K562cells was observed in the apoptosis induced by DADS; from the level of transcription,reverse transcription polymerse chain reaction(RT-PCR) technical was employed todetect the expression of caspase3mRNA and c-jun mRNA in apoptosis induced byDADS in K562cells; Lastly, immunohistochemical(IHC) staining was used to detectthe expression of phospho-c-jun(ser73) protein and caspase3protein in the apoptosisinduced by DADS.Results:1)after exposure to DADS3.0h, morphological change was observed inpartly K562cells, furthermore, the time of treatment with DADS is longer, themorphological change is more obvious. Compared with control group, its manifestati-on was smaller size, bright weaken and the edge of cell was obscure, more, thequantity of cell was fewer;2) after exposure to DADS3.0h, the expression ofcaspase3mRNA was enhanced gradually(p<0.05), it indicates DADS inducesapoptosis through up-regulation the expression of caspase3; In the earlier time ofDADS treatment(0.5h,1.0h), which apoptosis was not obvious, the level of c-jun mRNA was increased rapidly(p<0.05), however, after treatment with DADS3h, itdrops into the basal level which was same to control group (p>0.05);3) Move forward,we examined the expression of phospho-c-jun(ser73) protein and caspase3protein inapoptosis by immunohistochemical(IHC) staining morphometric quantitative analysis,the result of IHC shows, after treating with DADS24h, the average optical density ofphospho-c-jun and caspase3protein is up-regulation.(p<0.01).Conclusion: DADS induces K562cells apoptosis through up-regulation theexpression of phospho-c-jun(ser73) and caspase3.
