AO/EB stain demonstrated that treatment of40mg/L DADS with K562cellsfor48h showed the characteristic morphological changes of apoptosis.DNA gel electrophoresis revealed that treatment of40mg/L DADS with K562cells for48h could produce an obvious DNA ladder.3.RT-PCR results exhibited that treatment of40mg/L DADS with K562cells for48h led to an increased Bax mRNA Expression and a decreased Bag-1mRNAexpression compared with the control group(p<0.05).Conclusion1. DADS can induce apoptosis of human leukemia K562cells, and the optimalconcentration is40mg/L. DADS has apoptosis-inducing effect on K562cell in vitro;2. The related molecular mechanism is that DADS can activate the mitochondriaapoptosis way through upregulate Bax expression and downregulate Bag-1expression. | | Keywords/Search Tags: | Diallyl disulfide, K562cells, Apoptosis, Bax, Bag-1 | PDF Full Text Request | Related items |
