ObjectiveThe aim of the study was to explore the brain-gut mechanism of Qizhiweitong granules on functional dyspepsia(FD)by animal experiments.MethodsRats were randomly divided into 6 groups,including high-dose Qizhiweitong granules group,middle-dose Qizhiweitong granules group,low-dose Qizhiweitong granules,deanxit group,model group and control group.Except for the control group,the rats of other groups were conducted tail pain stimulation for 7 days to establish FD model.General condition of the rats was observed,such as weight,temperament,coat color,and food intake.Then each group was given the corresponding treatment for 14 days.The gastric emptying and intestinal propulsion were researched by labelling method and weighting method.The pathological manifestations of antrum were observed by HE staining.The level of 5-HT,SP,VIP,iNOS,CGRP in the medulla oblongata and antrum were detected by ELISA.The positive expression of 5-HT1 A receptor in the medulla oblongata was detected by immunohistochemistry.ResultsBefore treatment,food intake and weight of FD rats were significantly reduced compared with those of control group,and coat color of FD rats was pale.Meanwhile,the FD rats were anxious and tense compared with the control.After treatment,the weight and food intake were all increased in these treatment groups.Compared with model group,gastric emptying and intestinal propulsion in Qizhiweitong granules group and deanxit group were not significantly changed(P> 0.05).The 5-HT level in medulla oblongata and antrum in Qizhiweitong granules group and deanxit group were all increased compared with model group(P < 0.05).There was no significant change on the level of SP in medulla oblongata and antrum in Qizhiweitong granules group and deanxit group compared with model group(P > 0.05).The VIP level in medulla oblongata and antrum in Qizhiweitong granules group and deanxit group were not significantly different compared with model group(P > 0.05).The CGRP level in medulla oblongata and antrum in high-dose Qizhiweitong granules group and the middle-dose Qizhiweitong granules group were significantly reduced compared with model group(P < 0.05).And the CGRP level in high-dose Qizhiweitong granules group and the middle-dose Qizhiweitong granules group were significantly reduced compared with low-dose Qizhiweitong granules group(P < 0.05).The iNOS level in medulla oblongata and antrum in Qizhiweitong granules group and the deanxit group were not significantly different compared with model group(P > 0.05).Compared with model group,the expression of 5-HT1 A receptor in medulla oblongata in Qizhiweitong granules group and deanxit group were obvious increased(P < 0.05).ConclusionQizhiweitong granules may treat FD by reducing gastrointestinal high sensitivity by regulate the level of 5-HT,CGRP in medulla oblongata and antrum and 5-HT1 A receptor in medulla oblongata.
