| Objective:By detection of YAP,TAZ and β-catenin protein in normal gastric tissues,gastric mucosa of intraepithelial neoplasia tissues and in gastric cancer tissues,to explore the YAP,TAZ and β-catenin’s role and clinical significance of gastric cancer.Methods:1.Immunohistochemistry(IHC)was used to detect the expression of YAP,TAZ,β-catenin protein in 27 cases of normal gastric tissues,32 cases of gastric mucosa of intraepithelial neoplasia tissues and 87 cases of gastric carcinoma.Follow-up gastric cancer cases and analysis the clinical pathological parameters of YAP,TAZ and β-catenin protein.Investigate the relationship of these expressions.2.Western Blot was used to detect the expression of YAP,TAZ and β-catenin protein in 20 cases of fresh gastric cancer tissues and adjacent para-carcinoma tissues.Results:1.IHC results showed that YAP in normal gastric mucosa almost no expression,in gastric mucosa of intraepithelial neoplasia and gastric carcinoma nucleus was positive,intraepithelial neoplasia and gastric cancer mucosa tissue positive rate were higher than normal gastric mucosa(P<0.05),gastric intraepithelial neoplasia compared with normal gastric mucosa no statistical difference(P>0.05).TAZ in normal gastric mucosa almost no expression,in gastric mucosa of intraepithelial neoplasia and gastric cancer were nucleus positive,expression positive rate were higher than in normal gastric mucosa(P<0.05),and gastric mucosal intraepithelial neoplasia was higher than normal gastric mucosa(P<0.05).β-catenin expression in gastric mucosa intraepithelial neoplasia andgastric cancer was nucleus or cytoplasm positive,and positive rate was higher than normal gastric mucosa(P<0.05),gastric mucosal intraepithelial neoplasia was higher than the normal gastric mucosa(P<0.05).2.The expression of YAP was positively correlated with the expression of β-catenin(r=0.246,P=0.037).The expression of TAZ and β-catenin in gastric cancer was positively correlation(r=0.498,P=0.024).3.The expression of YAP protein was associated with lymph node metastasis and TNM stage(P<0.05)in patients with gastric cancer,and was not associated with age,sex and Lauren type(P>0.05).The expression of TAZ protein was associated with lymph node metastasis and TNM stage(P<0.05),age,gender,Lauren types were independent(P>0.05).β-catenin protein in patients with gastric cancer,Lauren type,lymph node metastasis and TNM stage were related(P<0.05),but the patient’s age,gender were independent(P>0.05).4.Gastric patients with YAP,TAZ or β-catenin abnormal expression,the survival time was shorter,the survival rate was lower(P<0.05).5.YAP,TAZ or β-catenin positive expression was risk factor for poor prognosis in patients with gastric cancer.6.Western Blot showed that the expressions of YAP,TAZ,β-catenin protein in gastric cancer tissues were significantly higher than that in adjacent tissues(P<0.05).Conclusion:YAP,TAZ and β-catenin played important roles in the occurrence and development of gastric cancer,the expressions level of these were important to determine the prognosis of gastric cancer. |
