Effects Of Periodontal Interventions On Carotid Artery And Serum Interleukin 6 In Periodontitis Rats Associated With Type 2 Diabetes Mellitus

Objective To explore the effect of Periodontal interventions on vascular disease in SD rats with chronic periodontitis associated with or without T2 DM by observing pathological change of carotid artery and detecting the levels of interleukin 6.Methods Forty-four male SD rats were randomly divided into four groups,group A(normal control),group B(Type 2 Diabetes Mellitus,T2DM)and group C(chronic periodontitis),group D(periodontitis associated with type 2 Diabetes Mellitus,CP+T2DM).Group C/D was further randomly divided into group C1/D1(natural process,seven)and group C2/D2(periodontal interventions therapy,seven).The Serum levels of IL-6 was detected in five different time points(T1~T5)by ELISA.The Pathological lesion of carotid artery plaque was stained with hematoxylinand eosin(HE).Results 1 Pathological results showed that the carotid artery of group A was normal;Besides thickened blood vessel walls,increased inflammatory cells,foam cells and disordered elastic fibers were found in vascular wall of group B;Irregular vessel wall were founded in group C/D,inflammatory cells and foam cells were gathered in group C1/D1.Besides elastic fibers were disordered and even broken in the groups,and the change in group D1 was greater.While in group C2/D2 few inflammatory cells and foam cells were found diffused distribution,and the walls of carotid artery blood vessel were different.and the condition of carotid artery blood vessel was greatly improved in group C2.2 The levels of serum IL-6 in group B became higher gradually(P<0.001),While the changes in group A were mild(P>0.05).As time went on,The levels of serum IL-6 in group C1 and group D1 were significantly higher at five time points(P<0.01),the level of IL-6 in each group reached respectively(C1/D1:337.985±35.866/487.724±18.831,pg·L-1)at the fifth time point;After the first interventions,the levels of serum IL-6 in groupD1 were significantly higher than group C1(P<0.001);At the first interventions,the levels of serum IL-6 in group C2 and Group D2 were significantly higher,reached respectively(C2/D2: 290.212±20.218/656.802±54.232,pg·L-1),then the levels declined with varying degrees at the third time point(P<0.01).Then the levels of serum IL-6 in periodontal interventions group C2 and group D2 were lower than that in group C1 and group D1 after the interventions had done(P<0.01,except the third time point).Conclusions 1 IL-6 may be involved in the development of CP and T2 DM,and was associated with vasculopathy 2 If no treatment,chronic periodontitis may accelerate the severity and the risk of vasculopathy in chronic periodontitis associated with or without T2DM;3 Periodontal interventions may increased inflammatory response in a short time,which may rise the risk of the development of vasculopathy in carotid artery,but may improve it after a long period of time;...
