| Objective:Type 2 diabetes mellitus,as a chronic disease,is a serious threat to human health.Some studies had shown beneficial effect of inulin on the blood glucose and lipid profiles.However,there is still controversy about the effects of inulin on glucose and lipid metabolism,so this study was designed to evaluate the effects of inulin on glycemic status and lipid profile in type 2 diabetic rats and discuss its possible mechanisms.Methods:Twenty-three male Sprague-Dawley(SD)rats were fed with standard diet for one week and according to the body weights,the rats were divided into normal control group(group A,n=8)and diabetic model preparation group(n=15).Group A: normal control rats(fed with standard diet),diabetic model preparation group rats(fed with high glucose and high fat diet).Diabetic model preparation group rats were injected with STZ after 4weeks to induce type 2 diabetic model rats.Twelve diabetic rats induced successfully were randomly divided into diabetic model(group B,n=5),diabetic rats treated with inulin(group C,n=7).Rats in group C were received inulin(3 g/kg/day).Measured the weekly fasting blood glucose and weight.All rats were sacrificed after taken blood at 14 weeks when inulin treatment was finished.After the rats were sacrificed,the adipose tissue was extracted.Plasma adiponectin,leptin and GLP-1 levels were assayed by Enzyme-linked immunosorbet assay(ELISA).Results:1.General characteristics in each groupThe symptoms of type 2 diabetes were obvious in the diabetic model group,such as dispirited,dull hair color,polydipsia,polyphagia,polyuria,and so on.Compared with model group,inulin group’s diabetes symptoms were lighter.2.Fasting blood glucose and lipid profile in each groupCompared with normal control rats,group B had significant increased fasting blood glucose(FBG),total cholesterol(TC),triglyceride(TG)and low density lipoprotein cholesterol(LDL-C)(P<0.05)and decreased in high density lipoprotein cholesterol(HDL-C)values(P<0.05).Compared with diabetic model rats,group C had significant decreased fasting blood glucose(FBG),total cholesterol(TC),triglyceride(TG)and low density lipoprotein cholesterol(LDL-C)(P<0.05)and increased in high density lipoprotein cholesterol(HDL-C)values(P<0.05).3.Body weight,white adipose tissue weight,brown adipose tissue weight,the ratio of white adipose tissue weight and the ratio of brown adipose tissue weight to body weightCompared with normal control rats,body weight,brown adipose tissue weight and the ratio of brown adipose tissue weight to body weight were significantly decreased in group B(P<0.05),white adipose tissue weight and the ratio of white adipose tissue weight to body weight were significantly increased in group B(P<0.05).After 14 weeks of inulin intervention,compare with group B,white adipose tissue weight and the ratio of white adipose tissue weight to body weight were significantly decreased in group C(P<0.05),and brown adipose tissue weight and the ratio of brown adipose tissue weight to body weight were significantly increased in group C(P<0.05),but body weight showed no significant change.4.The levels of plasma adiponectin,leptin and GLP-1The levels of plasma adiponectin and GLP-1 were significantly decreased in group B compare to normal control rats,and the level of plasma leptin were significantly increased in group B compare to group A(P<0.05).After inulin treatment,comparedwith group B,the levels of plasma adiponectin and GLP-1 were significantly increased and the level of plasma leptin were significantly decreased in group C(P<0.05).Conclusion:1.Continuous high sugar and fat fed and small doses of STZ intraperitoneal injection can almost copy type 2 diabetic rats model,it would be available to study T2 DM.2.Compared with diabetic model rats,the blood glucose and hyperlipidemia were obviously improved after the intervention of inulin,which suggested that inulin can reduce blood glucose and lipid profile.3.In diabetic rats,white adipose tissue weight and the ratio of white adipose tissue weight to body weight were significantly increased,brown adipose tissue weight and the ratio of brown adipose tissue weight to body weight were obviously decreased,the level of plasma adiponectin and GLP-1 were significantly decreased,and the level of plasma leptin were significantly increased.It is suggested that adipose tissue are closely to the occurrence of type 2 diabetes.4.After inulin treatment,white adipose tissue weight and the ratio of white adipose tissue weight to body weight were significantly decreased,brown adipose tissue weight and the ratio of brown adipose tissue weight to body weight were obviously increased,the level of plasma adiponectin and GLP-1 were significantly increased,and the level of plasma leptin were significantly decreased.It seems that beneficial effects of inulin on glycemic and lipid profile mainly mediated by browning of white adipose tissue and the influence of adipocytokines and gut peptides. |
