| Objective:Coronin3 protein is one of vertebrate seven coronaviruses,highly conserved,with WD40 Repeated sequence that interacts with F-actin and Arp2 / 3 complexes,regulates the recombination of cell actin skeletons,and proteins associated with tumor invasion.The aim of this study was to explore the expression of Coronin3 protein in colorectal cancer tissues,adjacent tissues and liver metastases,and to analyze the correlation between Coronin3 protein and clinical and pathological factors.Methods:Collected and sorted from January 2010 to December 2015 treatment in Shanxi Province Cancer Hospital,confirmed by postoperative pathology for simultaneous or abnormal colorectal cancer liver metastases,surgery R0 resection of patients with clinical and pathological data.In addition to the merger of multiple transfer line palliative surgery,with severe medical diseases affect the long-term survival of patients,to meet the group conditions.The clinical data collected include: patient name,gender,age,complication,location of primary tumor,depth of invasion,general type,type of organization,degree of differentiation,number of positive lymph nodes,number of liver metastases,location,treatment,Whether combined with multiple metastases,postoperative treatment,follow-up long-term survival and causes of death.The expression of Coronin3 protein was determined by immunohistochemistry and RT-PCR at the protein level and messenger RNA level,and the clinical and pathological data were analyzed.The data were processed by SPSS17.0 statistical software package.The data were calculated by using the independent sample t test.The counting data were analyzed by chi-square test.The Cox regression model was used to analyze the single factor and multi-factor analysis.Graphpad prism Make a drawing.Results:1.In this small sample of colorectal cancer patients with liver metastases,73%(22/30) of patients more than 60 years of age,men slightly more than women,the primary tumor site and colon more common 70%(21/30),The general type of ulcer type is more common,not all in situ tumors are infiltrating the whole layer of tissue,1 case of tumor invasion of the superficial muscle that distant metastasis,the majority of patients with more than 4 positive lymph nodes(N2),6 cases without lymph node metastasis,the number of liver metastases to 3 less common,preoperative CEA,AFP no significant increase in the majority.2.The positive rate of Coronin3 protein was 63%(19/30)in colorectal cancer and 67%(20 / 30).The expression of Coronin3 protein in the adjacent tissues was 2.60 ± 1.38,the expression in the in situ tumors was 4.43 ± 1.34,and the expression in the liver metastases was 5.12 ± 2.32.The expression of Coronin3 protein mRNA in paracancerous tissue was 0.78 ± 0.12,1.43 ± 0.56 in in situ tumor and 1.52 ± 0.72 in liver metastatic tissue.Correlation analysis showed that Coronin3 protein expression in adjacent tissues was significantly lower than in situ carcinoma and liver metastases,but there was no significant difference between in situ carcinoma and liver metastases.3.The expression of the Coronin3 protein is related to the number of metastases,the size of the tumor,the number of lymph nodes,and the age and sex of the patient.Conclusion:1.Colorectal cancer with liver metastasis feasible at the same time surgical resection of patients with colorectal cancer more common,the number of liver metastases is less than 3,the general type of pathology ulcer type more common.2.The expression of Coronin3 protein in the adjacent tissues was highly expressed in cancerous tissues and liver metastases,suggesting that it was related to the formation of tumor.3.The expression level of Coronin3 protein was correlated with the number of metastases,the number of positive lymph nodes and the degree of differentiation of primary tumor.However,there was no significant difference between cancer tissue and adjacent tissues.Suggesting that it is associated with the progression of colorectal tumors. |
