IntroductionThe early diagnosis of the malignant tumors as a first pivotal step determine the prognosis of patients.But present detection techniques which were used for clinical and screening,lack adequate sensitivity and specificity.SELDI TOF MS (surface-enhanced laser desorption/ionization time of flight mass spectrometry) is one of the recently developed sophisticated technologies, which, based on capturing proteins/peptides by chemically modified surface,is specifically powerfull for analyzing the complex biological samples. This technology,combined with bioinformatics,has been successfully used to analyze the complex serum proteins to explore the cancer-specific fingerprints or patterns.Material and Methods80 cases of the patients with colorectal cancer,41 cases of the healthy people and were tested by ProteinChip PBSIIC System of Ciphergen Inc.In second stage,proteomic spectra of 80 serum samples from patients with colorectal cancer and 41 from normal individuals were generated by CM-10 proteinchip array and SELDI-TOF mass spectroscopy. The discriminatory profiling between cancer and normal samples was analyzed by ProteinChip Software 3.2.1 and Biomarker Wizard.Results1 Between the 80 cases of the colorectal patient and 41 cases of the nomal control,we screened for 72 differentially expressed M/Z peaks. And we obtained 7 proteins(3961.44Da, 4286.32Da,6438.39Da, 3162.36Da, 4301.49Da, 4478.96Da, 6844.42 Da)making up markers pattern which was able to class the patient-team and normal-team. Corresponding accuracy ratio was 94.21%(114/121); sensitivity ratio was 92.50% (74/80); specificity ratio was 86.96%(40/46);2 Between the 27 cases of the colorectal patient with liver metastisis and 53 cases of no liver metastasis control,we screened for 6 differentially expressed M/Z peaks.3 Between the 20 cases of the colorectal patient with limphnode metastasis and 23 cases of no limphnode metastasis ,we screened for 5 differentially expressed M/Z peaks.Conclusions1.The protein marker group of which M/Z is 3961.44Da, 4286.32Da, 6438.39Da 3162.36Da in serum could be use to provide a highly accurate and innovative approach for the early diagnosis of colorectal cancer.2.The protein marker group of which M/Z is 5641.82Da 8703.02Da 6205.51Da 4353.81Da 3377.19Da 5907.43Da in serum could be use to provide a highly accurate and innovative approach to disti the liver metastasis patients and the non-liver metastasis patients,and to estimate the liver metastasis shoud occur or not in the future.3.The protein marker group of which M/Z is4628.80Da, 7777.21 Da, 4602.37 Da 9410.08 Da 4096.516 Da in serum could be use to predict the colorectal patients would occur limphnode metastasis or not.
