Objective:Select the depression modeling successful C57BL/6J model mice were grouped according to the type and dose of drug intervention,after the intervention,and the changes of serum IL-18 levels in brain in mice,correlation analysis of IL-18 level changes and the depression of the occurrence and development of disease,to explore the relationship between the level of peripheral blood IL-18 and incidence of depression,changes and immune mechanism the direction of change in the brain;to explore the level of serum IL-18 and IL-18 are the same;to explore changes in the progression of depression,as the possibility of a new objective biological index.Methods:According to a randomized method,60 healthy clean C57BL/6J mice model(purchased from the experimental animal center of Shanxi Medical University)in accordance with different dose and drug intervention,were randomly divided into 6 groups,10 rats in each group,respectively.The healthy control group,model control group,the Rositin depression treatment dose group and astragalus injection group.The high dose astragalus injection and Huangqi Injection in low dose group.Intervention: the intervention group according to the design of paroxetine dose of 5mg/kg/d orally,Huangqi Injection intervention group according to different doses of 0.025ml/kg/d,0.050ml/kg/d and0.075ml/kg/d respectively(corresponding to the original 4g/kg/d crude drug 2g/kg/d,and6g/kg/d)by intraperitoneal injection,a total of 56 days of intervention.There was no drug intervention in the control group,and the other treatments were the same as those in the intervention group.Before intervention,take the blood of mice,the depression model group after 56 days of the corresponding drug intervention groups again to take blood samples,and decapitated mice,removal of hippocampal tissue in mice,the paraffin sections after.ELISA was used to measure the level of serum IL-18 in mice,and the expression of IL-18 staining cells in hippocampus of mice was detected by immunohistochemistry.Finally,the data were analyzed by SPSS 22 statistical software.Results:1.Depression mouse model results After modeling 28 d,the mice were found to be prone to curl up,timid and frightened,hair rough,dull,body weight increased slowly,food intake reduced,and sugar consumption reduced;2.Serum IL-18 detection results(1)compared with the healthy control group,the serum IL-18 level of the model group was significantly higher than that of the healthy control group(P < 0.05);(2)after the intervention,the changes of depression model of depression model mice serum IL-18 level,Pa Rossi Dean group,high dose astragalus injection group and astragalus injection in the dose group serum IL-18 levels were significantly decreased,the difference was statistically significant(P < 0.05);and the levels of serum IL-18 in low dose group and saline group astragalus injection is not obvious,the difference was not statistically significant(P > 0.05);3.The results of IL-18 immunohistochemistry in hippocampus of mice The depression model mice after the intervention,the hippocampus stained cells were higher than that of control group,the difference was statistically significant(P < 0.05),the Pa Rossi Dean group,high dose astragalus injection group and astragalus injection in mice hippocampus stained cell number was significantly lower than that in normal saline group and low dose Astragalus injection group was statistically significant the difference(P <0.05).Conclusions:1.The change of serum IL-18 level may be related to the occurrence and development of depression.2.The changes of serum and hippocampus IL-18 levels in the serum of each group of depression model group,serum IL-18 levels can indirectly reflect the level of IL-18 in the brain.3,high dose of Astragalus injection and middle dose of Astragalus injection can play an effective antidepressant effect,low dose of Astragalus Injection antidepressant effect is not obvious.
