Objective: The purpose of this study was to explore the mechanism of sclerosing agent injecting CSA rabbit models with retrograde-tracing combining Immunohistochemistry method,as well as the effects of different lesion site on neck sympathetic nerve function.Methods: In part one,the rabbits were randomly divided into CSA group and control group.Transcranial Doppler(TCD)was used to detect the peak systolic velocity PSV),end diastolic velocity(EDV),mean velocity(Vm),pulsatile index(Pi),resistance index(RI)of the vertebral artery after the sclerosing agent was injected next to the right side parapophysis of cervical vertebra in the CSA group.In part two,rabbits were randomly divided into CSA group and control group,retrograde-tracing combining Immunohistochemistry method were used to detect the FB-positive neurons and the proportion of FB-positive neurons co-expressed neuropeptide Y(NPY)and tyrosine hydroxylase(TH)-immunoreactivity,western blot was used to detect the expressions of NPY and TH;In part three,rabbits were randomly divided into hypoderm group,upper trapezius muscle group and parapophysis group,retrograde-tracing combining Immunohistochemistry method were used to detect the FB-positive neurons and the proportion of FB-positive neurons co-expressed neuropeptide Y(NPY)and tyrosine hydroxylase(TH)-immunoreactivity,western blot was used to detect the expressions of NPY and TH.Result: Sclerosing agent established the CSA rabbit model successfully,and we found the TH and NPY increasing in this model,retrograde-tracing in different site and we found the FB-positive neurons in parapophysis group was more.Conclusion: Sclerosing agent established the CSA rabbit model successfully,and this model cause vertebral artery maybe through stimulating the sympathetic nerve;The distribution of sympathetic nerve at different site was different,this could be the reason of lesion in different site cause different effect.
