With coltsfoot leaf as raw material,extracting total flavonoids,total saponins and polysaccharide by extract,we get the optimum extraction conditions by using Design-Expert8.06 software designanalysis.And the total flavonoids,extraction of the total saponins and polysaccharide were purified and decolored,afterwards,the total flavonoids,total saponins and polysaccharide of coltsfoot were used to study the reduction of lipid,antioxidation and immunity.The results are as follows:1.Through the Box-Behnken analysis of Design-Expert8.06 software,the optimum extraction conditions of coltsfoot leaf flavonoids are: extracting time 30 min,the volume fraction of 80% ethanol,then(mL:g)liquid ratio,extract temperature 58 ?,the maximum withdrawal rate is 7.18%.After purification,the purity of total flavonoid of coltsfoot leaf was63.2%.2.Through the Box-Behnken analysis of Design-Expert8.06 software,the optimum extraction conditions of total saponins of coltsfoot leaf are: liquid ratio of land(mL: g),extracting temperature for 51 ?,extracting time for 13 min,extraction times 2 times,maximum Yield of 5.22%.After purification,the purity of coltsfoot leaf total saponin was60.4%.3.Through the Box-Behnken analysis of Design-Expert8.06 software,the optimum extraction conditions of total saponins of coltsfoot leaf are: : moreover(mL: g)ratio of liquid,extraction time is 34 min,extract temperature is 69 ?,3 extraction times,maximum Yield of1.94%.After decolorization,the purity of total flavonoid of coltsfoot leaf was 59.8%4.In reducing hematic fat test,we determined of mice serum TC,TG,LDL-C and HDL-C the content of Experimental,results show that flavonoid extract,saponin extracts and polysaccharides extract can reduce model mice TC,TG,LDL-C content in the body,increase HDL-C content,having a good cholesterol-lowering effect.Based on the analysis of the indexes,high flavonoids dose group has the best effect of reducing blood fat.5.In oxidation test,in mice liver tissue of T-SOD,MDA and GSH content were determined.Test results show that flavonoid extract,saponin extracts and polysaccharides extract can optimize the indicators,which showed good antioxidant effect.Comprehensive analysis on every index,the best antioxidant effect is yellow ketone group,followed by polysaccharidegroup,and at last the saponins group.6.Nonspecific immune test showed that the functional components such as total flavonoids total saponins and total polysaccharides in coltsfoot leaf can increase the carbon clearance ability of Immunosuppressed mice,the immune organ viscera coefficient,LZM content in serum and ACP activity,and ACP activity in liver tissue.Every effect of coltsfoot leaf components can improve the effect of immunity.Through the data analysis know that the best one is total polysaccharides,followed by total flavonoids,finally is total saponins.
