This paper studies on comparing two methods (Steam distillation and microwaveextraction) for essential oils extraction from the leaf of Platycladus orientalis. The microwaveextraction technology was optimized by response surface methodology based on single factorexperiment, meanwhile, the key factors of affecting essential oil extraction rate werediscussed by computer generated response surfaces and contour plot, and determine theoptimum technology for essential oils extraction from the leaf of Platycladus orientalis bymicrowave extraction. The extracted essential oils by two methods was analyzed for chemicalcomposition by GC-MS. The result that:1.steam distillation of optimal extract time was3h. extraction rate is1.3%.2. optimal values of extraction power, time and ratio of liquid/solid to material were1800W,30min and20/1, respectively. The advantages of this microwave extraction processrelative to the traditional steam distillation effectively shorten the extraction time, greatlyimproving the essential oil extraction rate (over2.72%) compounds extracted speciesincreased significantly (over8compounds).3.38compounds were isolated from essential oil, the component spectra obtainedNIST2005spectrum library retrieval by computer, and with reference to a standard spectraand mass spectrum charecteristic information, the38compounds were identified. essential oilby steam distillation27kinds of compounds, the total ion current figure of81.9%,careen39.79%,isolimonene6.86%,caryophelene5.32%,Cyclohexene-1-methanol,a,a,4-trimethyl acetate5.09%。4.microwave extraction method of the essential oil was isolated from compounds totalion current figure of99.41%,pinene34.16%,2-carene18.30%,limonene7.57%,caryophelen7.49%,terpinyl acetate5.36%,phellandrene3.69%.
