The Expression And Meaning Of AEG-1 And LEF-1 In Ocular Adnexal MALT Lymphoma

Objective: To explore the expression of AEG-1 and LEF-1 in ocular adnexal lymphoma,compared with reactive lymphoid hyperplasia.The relation between the expression of AEG-1 and LEF-1 and clinical pathological parameters,interrelation of AEG-1 and LEF-1,and how they affect the tumorigenesis of ocular adnexal lymphoma will be discussed.Method: Immunohistochemistry was used for evaluating the protein expression of AEG-1and LEF-1 on 40 samples of ocular adnexal lymphoma tissues and 20 reactive lymphoid hyperplasia specimens.In addition,AEG-1 and LEF-1 expression differentiation among clinical pathological parameters,including ages,gender,tumor occurrent site and pathological type,was analysed.Real-time Polymerase Chain Reaction(PCR)was used to test the AEG-1 and LEF-1 m RNA of 8 ocular adnexal lymphomas and 8 reactive lymphoid hyperplasia.Results: 1.AEG-1 protein expression and m RNA: a.AEG-1 positive granule mainly located in cytomembrane,less in cytoplasm(Figure 3).b.ratio of AEG-1 positive expression was62.5% in ocular adnexal MALT wich was higher than in eactive lymphoid hyperplasia of20%(P<0.05,Table 1).c.difference of AEG-1 protein expressions in varoius Ann Arbor clinical stage shows a statistical meaning(P<0.05,Table 2),but no relation when it comes to the age,gender,or occurent site.d.AEG-1 m RNA of ocular adnexal MALT was higher than that of reactive lymphoid hyperplasia(P<0.05,Figure 9).2.LEF-1 protein expression and m RNA: a.LEF-1 positive particles gather in cell nucleus(Figure 6).b.The percentage of LEF-1 positive expression in ocular adnexal MALT was higher than that of reactive lymphoid hyperplasia(P<0.05,Table 3).c.difference of LEF-1 protein expression in varoius Ann Arbor clinical stage shows a statistical meaning(P<0.05,Table 4),but no relation when it coms to the age,gender,or occurent site.d.LEF-1m RNA of ocular adnexal MALT was more than that of reactive lymphoid hyperplasia(P<0.05,Figure 10)3.The express correlation of AEG-1 and LEF-1: A positive revelance between AEG-1and LEF-1 was oberserved in ocular adnexal MALT(Table 5).Conclusion: AEG-1 and LEF-1 overexpression participate the development of ocular adnexal lymphoma.Their high expression closely relate to Ann Arbor clinical stage.The two factors may be a index parameter for diagnostic of ocular adnexal lymphoma,which will be useful for pathological diagnosis ocular adnexal MALT.
