Objective : To detect the effects of Roux-en-Y gastric bypass surgery on metabolic changes of hyperuricemia rats and investigate the underlying mechanism.Method : Forty male Wistar rats were randomized into the blank control group(C,n=10)and model group(M,n=30).The rats in model group were administrated with adenine100mg/(kg*d)by gavage,yeast-rich forage and potassium oxonate injection for 3 weeks.Then divided the model group into hyperuricemia group(H,n=10),sham operation group(S,n=10)and Roux-en-Y gastric bypass group(R,n=10)randomly.The rats in the R group underwent RYGB surgery,the rats in the S group underwent the operation in which gastric wall and jejunum were cut and then sutured in the original locations,and the rats in the H group were normal feed.Blooduricacid(UA),creatinine(CR),ureanitrogen(BUN),Triglycerides(TG),cholesterol(TC),fasting plasma glucose(FPG),fasting insulin(Fins),homeostatic model assessment of insulin resistance(HOMA-IR)were tested before operation and 2,4,6,8 weeks after operation to analys metabolic change.Results: Before surgery,the body weight of rats in each group had no statistical difference(p>0.05).After surgery,the body weight of RYGB group was significantly lower than other three group(p<0.05).Eight weeks after operation,the UA level of the RYGB group was significantly lower than that before operation(P<0.05),and also lower than that of S group and H groups in the same period(P<0.05);but had no statistical difference with that of the blank control group(P>0.05).The results were same to CR and BUN.The CR and BUN level of the RYGB group was significantly lower than that before operation(P<0.05),and also lower than that of S group and H groups in the same period(P<0.05);and had no statistical difference with that of the blank control group(P> 0.05).The TC level of the RYGB group was significantly lower than that before operation(P<0.05),and also lower than that of S group and H groups in the same period(P<0.05);and had no statistical difference with that of the blank control group(P>0.05).The TG level of the RYGB group,H group and S group were all significantly lower than that before operation(P<0.05),and no signifencant differences between each other(P>0.05),and had no statistical difference with that of the blank control group(P>0.05).For FPG,the level of the RYGB group was significantly lower than that before operation(P<0.05),and also lower than that of S group and H groups in the same period(P<0.05);and had no statistical difference with that of the blank control group(P>0.05).The Fins level and HOMA-IR level of the RYGB group was significantly lower than that before operation(P<0.05),and also lower than that of S group and H groups in the same period(P<0.05);and had no statistical difference with that of the blank control group(P>0.05).Conclusion: Roux-en-Y gastric bypass surgery can improve metabolic disorders of hyperuricemia rats.It may be related with the changes of gut hormones and microbiotawhich influence metabolic and inflammatory pathway.
