PartⅠ:The Establishment of SD Rat Roux-en-Y GastricBypass ModelObjective Establish the Roux-en-Y gastric bypass SD rat experiment model so as toprepare for the further establishment of diabetic rat model Roux-en-Y bypass model andstudy its hypoglycemic mechanism.Method Conduct RYGB on10normal8-week-old SD rat, establish a reasonablepreoperative, intraoperative, and postoperative operations as well as daily care methods,also10rats from sham-operated control group are used as compare group, and compare thechanges in weight1,2,3,4,5,6weeks before and after the surgery.Result The success rate of RYGB modeling is70%; compared with thesham-operated control group, differences in weights appeared in the3rdweek. Until the6thweek, RYGB can effectively reduce the weight and maintain for a long term.Conclusion The establishment of Roux-en-Y gastric bypass animal model throughSD rat is feasible. Roux-en-Y gastric bypass can effectively control the weight of SD rat. PartⅡ: Changes in Type2diabetes mellitus SD Rat Incretinafter Roux-en-Y SurgeryObjective Investigate the degradation mechanism of Roux-en-Y gastric bypass(RYGB) on the type2diabetes mellitus (T2DM) rats.Method406-week old male SD rats are fed by high-energy feedstuff combined withlow dose of streptozotocin (STZ,60mg/kg, intraperitoneal injection) to construct type2diabetes mellitus (T2DM) model. The32successfully modelled T2DM rats are randomlygrouped into normal comparison group (Group C), operative group (Group O),sham-operated control group (Group S), and food control group (Group F),8in each group.Inspect the changes in Fasting glucose, Serum insulin (FINS), Glucose-dependentinsulinotropic peptide (GIP), and Glucagon-like peptide-1(GLP-1)1,2,4, and8W beforeand after the surgery. Result Compared with C, S, F group, weight reduction took place in Group O1Wafter the surgery (P<0.5), Fasting glucose reduced (P<0.01), Glucose-dependentinsulinotropic peptide (GIP) decreases (P<0.05), and Glucagon-like peptide-1(GLP-1)increases (P<0.05)1,2,4, and8W before and after the surgery.Conclusion Roux-en-Y gastric bypass can significantly reduce the blood glucosevalue, its mechanism may be related with the secretion regulation. PartⅢ: The Impact of Roux-en-Y Gastric Bypass Surgery on Type2diabetesmellituss SD Rat Intestinal L CsObjective Explore the impact of Roux-en-Y gastric bypass (RYGB) on variousintestinal of type2diabetes mellitus (T2DM) rats.Method226-week old male SD rats are fed by high-energy feedstuff combined withlow dose of streptozotocin (STZ,60mg/kg, intraperitoneal injection) to construct type2diabetes mellitus (T2DM) model. The20successfully modelled T2DM rats are randomlygrouped into operative group (Group RYGB) and sham-operated control group (Group S),10rats in each group. Put the rats to death12weeks after the operation, and intestines ofRoux branch, pancreaticobiliary branch and common branches were taken respectively forimmunohistochemistry so as to calculate the density of L cells inside unit intestinalmucosa.Result Compared with Group S, weight and blood glucose of rats from Group RYGBdecreases significantly which maintained low weight and blood glucose for a long-term(P<0.05). RYGB significantly increases the area of upper intestinal mucosa of both Rouxbranch and common branch, as well as the number of intestinal L cells, but does notincrease the density of L cell (P>0.05), also there is no impact on pancreaticobiliarybranch.Conclusion Roux-en-Y gastric bypass can increase the area of intestinal mucosa andnumber of L cells, but cannot increase the density of L cell.
