Study On The Effect Of Different Combination Of Compound Lizards Powder Over The Expression Of Serum IL-4,Colon Tissue COX-2,MMP-9 In Rats With Ulcerative Colitis

Objectives Through animal experiment,to observe the influence of different combinations of compound lizard powders(XY80 mesh +100 mesh)on the content of serum IL-4,the expression of colon tissue COX-2,MMP-9 in rats with ulcerative colitis(UC),to reveal the possible mechanism of the treatment of UC.Methods 84 SPF male SD rats were fed in the experimental animal center for about 7 days and randomized into normal control group(group NC),model control group(group MC),sulfasalazine group(group SASP),compound lizards powder 80 mesh treatment group(group XY80 mesh),compound lizards powder 100 mesh treatment group(group XY100 mesh)and compound lizards powder 80 mesh +100 mesh mixed group(group XY80 mesh+100 mesh),a total of 6 groups,with 14 rats in each group.NC group rats were treated with saline enema,in the other groups,UC model was induced by the combination of three nitrobenzene sulfonic acid(TNBS)-ethanol complex method.After HE staining was used to determine the success of the model,the rats in each group were treated with the corresponding drugs for 14 days,the changes of general condition of rats were observed and DAI score was observed.At the end of the fourteenth day,all rats were fasted 24 h.After anesthesia and death,under the light microscope,to observe the pathological changes of rat colon tissue,enzyme linked immunosorbent assay(ELISA)detected the content of serum IL-4 and the expression level of COX-2 and MMP-9 in colon tissue was detected by immunohistochemistry,the data were analyzed by statistical methods.Results1.General situation: In the course of the experiment,we found that NC group rats were in good mental state and responsive,clothing hair is spic and span gloss,weight increased quickly,stool was normal;in the MC group,the mental state was found to be poor and the response was slow,get together without activity,weight loss,significantly reduced food intake,clothing hair fluffy and messy,dark and gloomy,loosestool even with mucous bloody,perianal fur was fecal contamination,yellow urine after second days;compared with the treatment group and the MC group,the above performance was improved in varying degrees,mental state improved,hair color bright,responsive,like foraging,stool gradually formed,with XY80 mesh +100 mesh group as the best.2.The disease activity index(DAI)score: The DAI score of each treatment group was significantly lower than that of MC group,which was statistically significant(P<0.05);the DAI score of SASP group,XY80 mesh group and XY100 mesh group were significantly higher than that of XY80 mesh +100 mesh group,which was statistically significant(P<0.05);there was no significant difference in DAI score between XY80 mesh group,XY100 mesh group and SASP group(P>0.05).3.Histopathological changes: HE staining was observed in the microscope,NC group colonic mucosa intact,duplicature smooth surface to rule,gland arrangement;MC group disappeared in colonic mucosa of each layer of tissue structure,cell gap widened,some of the mucosa was severely damaged,the glands were destroyed,a large number of eosinophils,neutrophils and lymphocytes infiltrated into the mucosa and submucosa to the muscularis;each treatment group compared with the MC group,the pathological results of colonic mucosa were improved,XY80 mesh +100 mesh group is the best,the basic structure of each layer is clear,can be seen scattered inflammatory cells.4.Statistical result: The treatment group compared with MC group,the content of serum IL-4 was significantly increased,the expression level of colon tissue COX-2 and MMP-9 was significantly decreased,there was significant difference(P<0.05);XY80 mesh +100 mesh group compared with other groups,the content of serum IL-4 increased obviously,the expression level of colon tissue COX-2 and MMP-9 decreased obviously,there was significant difference(P<0.05);comparison between SASP group,XY80 mesh group and XY100 mesh group of three,there was no difference in the content of serum IL-4,the expression level of colon tissue COX-2 and MMP-9(P>0.05).According to Pearson correlation analysis,there was a positive correlation between the expression of COX-2 and MMP-9(P<0.05,correlation coefficient r=0.969).Conclusion1.Different combinations of compound lizard powders can improve the general situation of UC model rats and the damage of colon mucosa,and can reduce the DAI score.2.Different combinations of compound lizard powders can increase the content of IL-4 and decrease the expression level of COX-2 and MMP-9.3.The expression of COX-2 was positively related to the expression of MMP-9,indicating that COX-2 and MMP-9 have synergistic effects in the development and progression of UC.4.The unique formula of different combinations of compound lizard powders,the mechanism may be through increasing the content of anti-inflammatory factor IL-4,to interfere with the activation of NF-kappa B signaling pathway,reduce the expression of inflammatory related enzymes COX-2 and MMP-9 and inhibit the expression level of inflammatory mediators,regulate immune imbalance,reduce the inflammatory response of intestinal tract and promote the repair of damaged colonic mucosa.5.Compound lizards powder can enhance the therapeutic effect after the combination of different particles,the effect of the repair of colonic mucosal ulcer is significantly better than only using 80 mesh particles or only using 100 mesh particles.
