Objectives Through animal experiment observe the influence of compound lizard powders(XY80 mesh+100 mesh)on the content of serum IL-6,the expression of colon tissue PPAR?,P65,i NOS in rats with ulcerative colitis(UC),to use compound lizard powders treat UC provide experimental basis.Methods 84 SPF SD rats,aged from 4 to 6 weeks,weight 190±20g were fed in the experimental animal center for about 7 days and randomized into blank control group,model control group,sulfasalazine group(group SASP),compound lizards powder 80 mesh treatment group(group XY80 mesh),compound lizards powder 100 mesh treatment group(group XY100 mesh)and compound lizards powder 80mesh+100 mesh mixed group(group XY80 mesh+100 mesh),a total of 6 groups,with14 rats in each group.The six groups are numbered A ~ F.NC group rats were treated with saline enema,in the other five groups,was induced by the combination of three nitrobenzene sulfuric acid(TNBS)-ethanol complex method.The fourth day of the enema randomly selected a rat to kill for test,After HE staining was used to determine the success of the model,the rats in each group were treated with the corresponding drugs for 14 days,the changes of general condition of rats were observed,After the 15 th day of anesthesia,all rats were executed,under the microscope observed the pathological morphology of the colonic mucosa,enzyme linked immunosorbent assay(ELISA)detected the content of serum IL-6 and the expression level of PPAR-? ?NF-?Bp65and i NOS in colon tissue was detected by immunohistochemistry,the data were analyzed by statistical methods.Results1.The general behavior in rats:We have observed during the experiment,In t he blank group,the rats fur is white and shine color,and the body surfacetempera ture is normal,the water intake is normal,the body weight is increasing daily,the stool is granular and there is no pollution in perianal.In the m-odel control grou p,SASP group?FFXY80 group?FFXY100 group?FFXY80+100 group,rats have different degrees of pathological manifestations by dep-ression,poor response and like cluster,weight loss,food intake decreases,the color yellow fleeciness,defecate appears thin pond with mucous purulent blood,surrounded by dilute pond anus bowel movement,yellow c-olor of urine,etc.After treatment,other group compared with model group rats there are different degrees of improvement.The FFXY80+100 group rats condition is best.2.Pathological and histological changes:See to the naked eye:blank control group rats colon normal form without the shortening of the circuity,good stretch,in neat rows and dense,intestinal wall edema,erosion,hemorrhage;The model control group?SASP group?FFXY80 group?FFXY100 group?FFXY80+100 group had different degrees of tortuous shrinkage,poor stretch ability,brittleness change,intestinal stenos is,mucosal uplift and so on HE staining was observed in the microscope,blank control group colonic mucosa intact,duplicator smooth surface to rule,gland arrangement;model control group disappeared in colonic mucosa of each layer of tissue structure,cell gap widened,some of the mucosa was severely damaged,the glands were destroyed,a large number of eosinophils,neutrophils and lymphocytes infiltrated into the mucosa and sub mucosa to the muscular issue;each treatment group compared with the model control group,the pathological results of colonic mucosa were improved.3.ELISA results:Model group serum proinflammatory factor IL-6 levels increased significantly,Compared with group A,every group had significant difference(P<0.05);The treatment group compared with model group,Serum IL-6content is lower;the difference was statistically significant(P<0.05),But FFXY80 group compare with FFXY100 group,difference is insignificant.4.Immunohistochemical results:Each group compared with the blank group,the expression level of colon tissue PPAR-? was significantly decreased,compare was significant difference(P<0.05),Compared with model group,PPAR-? protein expression levels were elevated,compare was significant difference(P<0.05),FFXY80+100 group compare with SASP group?FFXY80 group?FFXY100 group PPAR-? protein expression levels were elevated,compare was significant difference(P<0.05),FFXY80 group and FFXY100 group compare,difference was not statistically significant(P>0.05).Compared with the blank group,Each group the expression level of colon tissue NF-?Bp65 and i NOS was reduce significantly,There are significant differences,The treatment group compared with model group,NF-?Bp65 and i NOS was significantly decreased,compare was significant difference(P<0.05),FFXY80+100group compare with SASP group?FFXY80 group?FFXY100 group NF-?Bp65 and i NOS protein expression levels were elevated,compare was significant difference(P<0.05),FFXY80 group and FFXY100 group compare,NF-?Bp65 and i NOS protein expression levels was not statistically significant(P>0.05).Conclusion1.The combinations of compound lizard powders can improve the general situation of UC model rats,could reduce the content of IL-6,increase the content of and decrease the expression level of NF-?B p65 and i NOS,elevated the expression level of PPAR-?.2.Compound lizard powder formulation is unique,After different granularity combination enhance effect,Role of mucous membrane have obvious repair,the effect of the repair of colonic mucosal ulcer is significantly better than only using80 mesh particles or only using 100 mesh particles.
